Polygonatum sibiricum is from Liliaceae's Polygonatum's Polygonatum sibiricum Red., Polygonatum kingianum Coll.et Hemsl., Polygonatum cyrtonema Hua.'s dry root medicine. These three Polygonatum sibiricum are pharmacopoeia of the People's Republic of China(the 2010 edition) included varieties. Polygonatum sibiricum Red.are distributed in Shaanxi, Hebei and others provinces. Polygonatum kingianum Coll.et Hemsl.are distributed in Guangxi, Yunnan and others provinces,Polygonatum cyrtonema Hua.are distributed in Hunan and Guizhou provinces. Polygonatum sibiricum Red.is useful to kidney and spleen from the "Treatise on Thunder Gun." Polygonatum sibiricum is one of our traditional Chinese medicine of more commonly used. Polygonatum sibiricum has been used to treat diabetes, anti-tumor, enhance immunity and other diseases now.In order to improve the quality standards of Polygonatum sibiricum medicine further, comparison of the suitable area for growth of Polygonatum sibiricum Red., as well as provide reference for the standardization of Hanzhong Lueyang base's Polygonatum sibiricum Red.in this study. In this reseach,eight batches of Polygonatum sibiricum Red.in Yangling,Shaanxi (eight different times since the excavation of Yangling, Shaanxi) high performance liquid chromatography(HPLC) fingerprint(hereinafter referred to as Polygonatum sibiricun Red.HPLC fingerprint) were studied, using the total average vector pattern generation method and the common pattern generation method established the standard fingerprint of 70% ethanol extract of Polygonatum sibiricum Red. fromYangling, Shaanxi., Also on the eight different places Polygonatum HPLC fingerprint were studied,with some of the main active ingredient index determination of Polygonatum quality standards to a certain extent to supplement and complete, expected to give Polygonatum quality standards provide a reference. The main contents and the results are as follows:1 Study on fingerprint of Polygonatum sibiricum Red. from Yangling,Shaanxi by High Performance Liquid Chromatography (HPLC).This study is on fingerprint of those Polygonatum sibiricum Red. which collected from different periods of Yangling, Shaanxi by HPLC. The chromatographic conditions were optimized: Waters 1525 binary HPLC, Waters 2996 diode array detector, Waters UV-2487 detector, Waters XB-C18 column (250mm×4.6mm, 5μm), Empower2 Chromatography Software. To A: acetonitrile and B: water as mobile phase, Using gradient elution, Gradient proceduresare: 0-10min, 5%-10%A(v/v). 10-30min, 10%-35%A(v/v). 30-40min, 35%-60%A(v/v). 40-60min, 60%-100%A(v/v). Flowing rate is 1mL/min, Column temperature is 30℃, Detection wavelength is 200nm, The injection volume is 20μL. The results show that the method established HPLC fingerprint of Polygonatum specific, accurate, reliable, To reflect more truly comprehensive Yangling Polygonatum quality.2 Study on eight different origin place Polygonatum sibiricum Red.by HPLC fingerprintsUsing the method which be used to established the standard HPLC fingerprint of Polygonatum sibiricum Red.from Yangling, Shaanxi to reaseach the HPLC fingerprint from eight different areas of Polygonatum sibiricum Red.. Based on eight different origins Polygonatum sibiricum Red.'s HPLC fingerprint, we found that the main active ingredients in eight different origins Polygonatum sibiricum Red. are same, but the same ingredient content take on differences. It also shows the origin of the quality difference between Polygonatum sibiricum Red. Also studied using HPLC to study on the content of diosgenin between the Polygonatum sibiricum Red.of different origin, the results show that content of diosgenin of Polygonatum sibiricum Red. from AnGuo, HeBei is the highest relatively, followed Hanzhong, Shaanxi, Puer, Yunnan is the lowest.3 Study on ash content of Polygonatum sibiricum Red.Studying eight different origins Polygonatum sibiricum Red.'s total ash and insoluble ash content, the results showed that Anguo, Hebei and Zunyi, Guizhou's acid insoluble ash was not detected and other six different origin Polygonatum sibiricum Red.'s two ash contents are below the provisions chinese pharmacopoeia, belong to eligible samples, and also found that Xinxiang, Henanin which the two are relatively high ash content, and Zunyi, Guizhou Polygonatum total ash content is relatively minimal, Hanzhong, Shaanxi's Polygonatum sibiricum Red.acid insoluble ash content of at least. This may be attribute to the process of transportation related storage and handling4 Study on extract content of Polygonatum sibiricum Red.According to latest edition chinese pharmacopoeia(2010) demand,study eight different origins Polygonatum sibiricum Red.'s extract content, the results should that: Polygonatum sibiricum Red.of Anguo Hebei province's alcohol-soluble extract get the highest levels, Hanzhong lveyang base as the second, Yangling,Shaanxi the third, and all of them conform to the chinese pharmacopoeia.5 Study on polysaccharide content of Polygonatum sibiricum Red.According to the China pharmacopoeia(2010), study eight different origins Polygonatum sibiricum Red.'s polysaccharide by uv s000pectrophotometry, the result shows: Hanzhong lveyang base polysaccharide content is the highest, Anguo, Hebei the second, Yangling, Shanxi polysaccharide also relatively high. The polysaccharides have many functions,such as: anti-tumor, antiphlogistic aseptic, enhance immunity, etc.So this kind of research results explained that beside the origin place, climate factors,the cultivation and production methods also have some effect on the quality of the traditional Chinese medicine improve.6 The variance Analysis of evaluation factor of Polygonatum sibiricum RedDPS software be used to research the water, total ash, extract, diosgenin, polysaccharides, the similarity assessment of two ways of fingerprint, as seven of during eight different areas to find quality factor analysis of variance.The results showed: diosgenin and polysaccharide during different areas significantly, indicating that these two indicators should be used to evaluate the quality of Polygonatum sibiricum Red., this article suggests that the two indicators and the HPLC fingerprint of Polygonatum sibiricum Red. quality standards as well as a reference .
