Objective: To seek the different proteins in DDP-sensitive ovarian cancer cell OV2008 and DDP-resistant ovarian cancer cell C13K by two-dimensional electrophoresis. Analyze these proteins by Mass Spectrum to find ovarian cancer biomarker and the mechanism of ovarian cancer chemoresistance.Measures: To seek the different proteins in DDP-sensitive ovarian cancer cell OV2008 and DDP-resistance one C13K by two-dimensional electrophoresis. By the mass spectrometry analyzed the targets and verified the targets by Real-time PCR and Western blot.Results: .We sought sixteen different proteins between OV2008 and C13K and chosen five targets to verify them by Real-time PCT and Western-blot. We found that TCTP,Stathmin1/Oncoprotein18,eIF-5A,HSP27 increased in DDP-resistant ovarian cancer cell C13K and ERP29 increased in DDP-sensitive ovarian cancer cell OV2008.Conclusion: Our established proteomic technique of ovarian cancer cells OV2008 and C13K can be effectively applied in comparative proteomics of relevant chemo-resistant proteins in ovarian cancer. Objective: Recent studies have shown that MACC1 (metastasis-associated in colon cancer-1) is related to carcinogenesis and development of colorectal cancer, meanwhile it can be useful for the diagnosis of colorectal cancer. However it has never been explored in ovarian cancers. This study dedicates to investigate the expression and clinical pathological significance of MACC1 in epithelial ovarian cancer.Measures: The expression of MACC1 was detected by immunohistochemistry technique in cancerous tissue from cases of EOC and cases of normal ovary/benign/borderline. Results: The expressions of MACC1 was higher in the epithelial ovarian cancer than that in the normal ovarian tissue ,the benign ovarian tumor and borderline tumor (P<0.05),it also expressed higher in the benign ovarian tumor and borderline tumor than in the normal ovarian tissue. However, the difference between the benign ovarian tumor and borderline tumor had no significance (P>0.05).Furthermore, The positive rate of MACC1 was evidently higher in the moderately- and poorly- differentiated ovarian cancer than in the well-differentiated cases (P<0.05), and the level was markedly lower in StagesⅠandⅠof ovarian cancer tissues than that in StagesⅢandⅢ(P<0.05 ). The positive incidence of MACC1 was statistically associated with tumor metastasis(P<0.05),which unrelated ages.Conclusion: MACC1 expression elevates in epithelial ovarian cancer which may regard as a prognostic factor in ovarian cancer and provide a potential in targeted molecular therapy.
