The Expression Of TCTP In Human Ovarian Cancer And The Study Of Effect On Invasion And Metastasis In Ovarian Cancer Cells

[Objective] To investigate the expression of TCTP in epithelial ovarian cancerand analyze the relationship TCTP and clinical pathological characteristics.[Methods] The expression of TCTP was examined byimmunohistochemistry(IHC) in normal ovarian tissues of18cases, benignovarian tumors of20casses and epithelial ovarian cancerous tissues of60cases.[Results] The immunohistochemical staining shew that the expression ofTCTP in epithelial ovarian cancerous tissues was higher than that in normalovarian tissues and benign ovarian tumors(P<0.05). However, the differencebetween normal ovarian tissues and benign ovarian tumors had nosignificance(P>0.05). The expression of TCTP was associated with clinicalsatge, histological grade and lymph node metastasis, but not age(P>0.05).[Conclusion] TCTP overexpression may promote the progress of malignantphenotype in epithelial ovarian cancer. [Objective] To explore the impact of TCTP in proliferation, invasion andmetastasis in human ovarian cancer cells.[Methods] The expression of TCTP was detected in human ovarian cancercells with Real-time PCR and Western Blot.Synthesized siRNA wastransfected into ovarian cancer cell line with high expression of TCTP, andReal-time PCR and Western Blot was used to detect efficiency of siRNA. Theproliferation of SKOV3change before and after TCTP knockdown weremeasured by CCK-8, the migration and invasion changes were detected bychamber assays.[Results] The expression of TCTP in SKOV3cells was hagher than that inA2780,C13K,OV2008cells.Down-regulated expression of TCTP couldsuppress the proliferation, migration and invasion of SKOV3cells.[Conclusion] Down-regulated of TCTP expression by siRNA could suppressmalignant phenotype in epithelial ovarian cancer.TCTP has the potential tobecome a new therapeutic target for ovarian cancer.
