Objective :To investigate the Function of Th17cells and CD4+ IL-10+ Treg cells of peripheral blood in the patients of allergic rhinitis and Desensitization treatment effective.Methods:10 patients of severe perennial allergic rhinitis(AR)due to mite,10 healthy controls(HC)andDesensitization treatment effective(DE) were enrolled.peripheral blood mononuclear cells(PBMCs) were collected, respectively divided into three groups: ionomycin group(blank group) ,adding ionomycin 1ug/ml and monesin 2ug/l; mite group, adding ionomycin 1ug/ml; mite allergen nderp1 1ug and monesin 2ug/ml; PMA group , adding PMA 20ng/ml; ionomycin 1ug/ml and monesin 2ugl/ml. Cells were activated for 6h in carbondioxide cell incubator under 37℃,5% CO2.Then it was continuous to do intracellular and membrane staining with PBMCs and survey the percent of th17 cell in CD4+ cell by flow cytometric detector, and analyze the function of Th17 cells in the patients of AR and DE. Probe the percent of IL-10 cell in CD4 t cell. Results: The percents of Th17 cell in HC, Mite and PMA groups were inferior to corresponding AR group(P<0.05), also the percents of th17 cell in DE and Mite were be inferior to corresponding AR group(P<0.05). After the stimulated by mite, it was compared to blank AR decreased(P<0.05. After the stimulated by PMA, the percent of Th17 cell in HC, AR and DE were higher than blank group(P<0.05=. Otherwise the Treg cell of IL-10+ in HC, DE were lower than AR (P<0.05=. After stimulated by mite, AR was decreased to blank on the Treg cell of IL-10+ (P<0.05).Conclusions: It suggested that Th17 cells maybe had an influence of AR.PMA could increase the percent of Th cells secreting IL-17. It demonstrated the Treg cell secreting il-10 has a role of immunologic mechanism on allergic rhinitis. After the stimulated by PMA, the percent of Th1 cell in HC, AR and DE were higher than blank group(P<0.05)Conclusions: It suggested that Th2 cells maybe had a special memory of AR.PMA could increase the percent of IFN-γand decrease the percent of IL-4.
