Objective: Investigation the relationship of prostate cancer and bone metastasis through detecting the content of uPA and uPAR in serum of benign prostatic hyperplasia patients and prostate canceras as well as correlations between uPA,uPAR and prostate volume, Gleason score and serum PSA. Discussion the value of clinical diagnosis and prognosis with the levels of uPA and uPAR in serum of prostate cancer patients. Methods: The serum specimens are divided into three groups:1, the gruop of benign prostatic hyperplasia (36 cases); 2, the group of prostate cancer with not bone metastasis (15cases); 3, the group of prostate cancer with bone metastases(21 cases). Detection the expression of uPA and uPAR in serum by double antibody sandwich ELISA (enzyme-linked immunosorbent assay, ELISA). All data was analyzed by SPSS 18.0 statistical software. Detective results represent with X±S and were used to process data with One-Way ANOVA,each group's multiple comparison with LSD test,and then do the relevant analysis to detect if there was correlation between them and prostate volume,Gleason score as well as T-PSA in patients with PCa.α=0.05 was regarded as the statistical criterion and P<0.001 was regarded to be with significant difference. Results: The expression levels of uPA and uPAR in group 3 is (1.2000±0.2311 ng / ml) and (4.5412±1.1398 ng/ml).They are ignificantly higher than that of group 2 is (0.9459±0.3058 ng / ml) and (3.2994±0.9928 ng/ml),P=0.003<0.05 and P=0.000<0.05. The expression levels of uPA and uPAR in group 2 are ignificantly higher than that of group 1 is (0.7372±0.2217 ng/ml) and (2.6640±0.7002 ng/ml),P=0.007<0.05 and P=0.026<0.05. Group 3 are ignificantly higher than group 1,P=0.000<0.05 and P=0.000<0.05. Both of those differences are statistical significance. There was no statistically significant correlation between serum uPA and prostate volume,Gleason score as well as T-PSA in patients with PCa after correlation test,and so was uPAR (P>0.05). Conclusions: 1. The serum level of uPA, uPAR in patients with BM PCa were significantly higher than patients with non-BM PCa. 2. The serum level of uPA,uPAR in patients with non-BM PCa were significantly higher than patients with BPH. 3. The over expression of serum uPA,uPAR in patients with PCa are related with PCa and bone metastasis. 4. There were no correlation be observed between the serum uPA,uPAR and the prostate volume as well as Gleason score and serum PSA in prostate cancer patients. 5. Detection of the levels of uPA and uPAR in serum may help to differentiate benign prostatic hyperplasia and prostate cancer and to understand the situation of metastasis of prostate cancer and to guide clinical diagnosis and treatment .
