Objective:To study the relation of the mRNA and protein expression of CyclinD1,CDK2,CDK4 in different stages hypertrophic scar fibroblast(FB) with its cell cycle,and provide theoretical evidence for intervention therapy of cell cycle gene being used in hypertrophic scar.Methods:The hyperplastic scar samples in 32 cases and the normal skin samples in 8 cases were used in this study.The expression of CyclinD1,CDK2,CDK4mRNA and protein were detected by quantitative real-time PCR and Wetern Blot.And at the same time,the flow cytometer was used to detect the fibroblastic cell life cycle.Results:(1)The expression mRNA and protein of CyclinD1 in 3 months group,6 months group, 1year group,2 years group respective were14.17±1.51,0.74±0.16; 13.30±1.12,0.71±0.08; 9.73±1.01,0.46±0.08; 9.09±0.97,0.43±0.05; The mRNA and protein of CDK2 were15.41±1.56,0.77±0.16; 15.14±1.12.0.73±0.07; 8.53±1.87,0.39±0.03; 7.98±0.77,0.35±0.04; The mRNA and protein of CDK4 were 19.71±2.04,1.95±0.21; 12.04±1.32,0.79±0.08; 7.90±1.27,0.32±0.0; 7.75±0.17,0.29±0.09。The expression intensity mRNA and protein of CyclinD1,CDK2 in 3 months group and 6 months group was no difference(P>0.05),and that of in 3 months group and 6 months group was higher respectively than that of 1years,2 years and normal group (P<0.05), and that of in 1 year group,2 years group and normal group were no difference (P>0.05). The expression intensity mRNA and protein of CDK4 in 3 months group was higher than that of 6 months.And that of in 3 months group and 6 months group was higher respectively than that of 1years,2 years and normal group (P<0.05), and that of in 1 year group,2 years group and normal group were no difference(P>0.05). (2) Results of flow cytometry:Most FB were in S and G2/M stage (cycle) in 3 months,6months group;And most FB were in G0/G1 stage (cycle) in 1 year,2 years group and normal group.Conclusion:Following hypertrophic scar development, the expression mRNA and protein of CyclinD1,CDK2,CDK4 in fibroblasts of hypertrophic scar experienced from high level to low leve.The mRNA and protein expressions of CyclinD1,CDK2,CDK4 in different stages of hypertrophic scar are basically consistent, And the distribution of fibroblastic in cell life cycle match to the function of CyclinD1,CDK2,CDK4;The high percentage of fibroblastic cells at S and G2/M in esrly stage of hypertrophic scar suggest that an intervening in early stage be an important target to the genesis and development of hypertrophic scar.
