Tumor is a kind of malignant illness that the cell cycle is disturbed and the cell growth is loss of control acted by hereditary and environment factors. The main reason of cell over hyperplasia is disturbed cell cycle control. Cyclin has important roll in cell cycle control. There are A, D1 and E type of cyclin, which have close relationship with tumor. Recently considered, CyclinDl can connect with P16 protein competitively, induce PRb to phosphorylate, make inhibition of PRb growth disappear, change static state of cell into hyperplasia state.Morbidity and mortality of colon carcinoma are very high in common alimentary channel malignant tumor. That was confirmed, the occurrence and development of colon carcinoma is a procedure regulated by multi-genes, activation of tumor gene and inactivation of tumor suppressor gene involved in cell cycle are important elements in cell malignant hyperplasia and metaplasia. More and more evidences show disturbed cell cycle control is a important mechanism of tumor occurrence, abnormal changes of every regulator in G1/S stage have the most close relation with cell canceration. Present research show P16,CyclinD1 and CDK4 are all important control genes of cyclin, when any reason lead the abnormal expression of those genes, the cell cycle will loss of control, the tumor will occur and gradual progress malignantly. Many reaches show the occurrence and development of the malignant alimentary channel tumor have close relation with those three factors. In this trial, we performed immunohistochemical staining for CyclinD1,CDK4 and P16 test the expression in colon carcinoma and discover the relationship between those and clinical index of colon carcinoma, and discover some experimental evidence of CyclinDl and CDK4 involved in occurrence and development of colon carcinoma. Objective:To study the expression of P16,CyclinD1 and CDK4 in colon carcinoma and discover the relation ship between those and colon carcinoma and analysis the implication and relationship between those and clinical index of colon carcinoma we performed immunohistochemical S-P staining in this reach.Methods:The expression of CDK4,CyclinD1 and P16 were determined by S-P immunohistochemical staining in 60 cases of tissue of colon carcinoma and 60 cases of tissue adjacent to colon carcinoma, and analysis the relationship between those and clinical index.Statistic Analysis:SPSS 14.0 statistic software was applied. All data were analyzed with X2 test and p<0.05 was regarded as statistically significance.Results:The expression rate of P16 in tissue adjacent to colon carcinoma is markly higher than in carcinoma, and which have relation with Duke's stage and differentiated stage (P<0.05); the expression rate of CDK4 in tissue of colon carcinoma is markly higher than in tissue adjacent to colon carcinoma. The higher Duke's stage, the lower differentiated stage of tissue of colon carcinoma, the positive rate of CDK4 expression is obviously higher (P<0.05); the expression rate of CyclinDl in tissue of colon carcinoma is markly higher than in tissue adjacent to colon carcinoma, and the higher Duke's stage, the lower differentiated stage of cancerous tissue, the expression of CyclinDl is up-regulation apparently (P<0.05); the expression of P16,CyclinDl and CDK4 in tissue of colon carcinoma, CyclinD1 and CDK4 are over expressed and expression of P16 protein is decrease, after related analysis the study result, the positive rate of CDK4 and CyclinDl expression in tissue of colon carcinoma is conforming; The positive rate of P16 expression in colonic cancerous tissue with lymph node metastasis is lower than that in colonic cancerous tissue without lymph node metastasis (P<0.01), the positive rate of CyclinDl and CDK4 expression in colonic cancerous tissue with lymph node metastasis is higher than that in colonic cancerous tissue without lymph node metastasis (P<0.05).Conclusion:1. P16 is commonly expressed in cell plasma and nucleolus the positive rate of P16 expression in tissue of colon carcinoma is much lower than in tissue adjacent to colon carcinoma; with the higher Duke s stage, the lower differentiated stage of cancerous tissue, the deeper invasive depth of tissue, the positive rate of P16 expression in tissue of colon cancer with lymph node metastasis is lower than in tissue of colon carcinoma without lymph node metastasis.2. CDK4 and CyclinDl is commonly expressed in nucleolus, expression rate of CDK4 and CyclinDl in tissue adjacent to colon carcinoma is lower, however expression rate of CDK4 and CyclinD1 in tissue of colon carcinoma is higher, with the higher Duke's stage, the higher malignant degree, the lower differentiated stage of cancerous tissue, the expression is up-regulation, the positive rate of CDK4 and CyclinD1 expression in tissue of colon cancer with lymph node metastasis is higher than in tissue of colon carcinoma without lymph node metastasis.Down regulation of P16 expression in tissue of colon carcinoma and up regulation of CyclinD1 and CDK4 expression have important role in occurrence and development of colon carcinoma, and can be recognized as the malignant degree marker of colon carcinoma.
