| Objective:TO analy the correlation between HMGB1 expressions and the percentages of pancreatic acini oncosis in the rats with severe acute pancreatitis and to explore the mechanism of oncosis induced by HMGB1.Method:80 healthy SD rats were randomly divided into four groups:sham operation group (A group, n=8), SAP model group (B group, n=24), and PDTC-treated group(C groun=24),PDTC+EP treated group(D group, n=24). The sham operation group were only flipped the intestinal canal; the SAPmodel group were induced by retrograde injection of 3% sodium taurocholate into bilio-pancreatic duct; the PDTC-treated group were administrated with injection of PDTC before retrograde injection of 3% sodium taurocholate; PDTC+EP treated group received PDTC and EP by tail vein beginning. All rats were sacrificed after SAP had been induced at 6 hours,12 hours and 24 hours. Serumamy-HMGB1 expressions were measured by ELISA.; the percentages of pancreatic acini oncosis were examined by flow cytometry;the NF-κB levels were measured by Immnohistochemistry.While we analyzed the correlation between HMGB1 expressions and the percentages of pancreatic acini oncosis; the correlation between NF-κB levels and the percentages of pancreatic acini oncosis; the correlation between HMGB1 expressions and NF-κB levels.Results:The result showed HMGB1 expressions and the percentages of pancreatic acini oncosis in GroupB significant increased compared with that in GroupA at 6 hours,12 hours and 24 hours (P=0.000). There was positive correlation between HMGB1 expressions and the percentages of pancreatic acini oncosis (r=0.846,P=0.000). Compared with Group A the NF-κB levels in GroupB increased at 6hours,12hours and 24 hours(P=0.000),specially the NF-κB level at12 hours was higher than at 6 hours(P=0.015) in GroupB, but no difference at 24 hours(P=0.806). There was positive correlation between NF-κB levels and the percentages of pancreatic acini oncosis (r=0.731,P=0.000) in GroupB in 24 hours. There was positive correlation between HMGB1 expressions andNF-κB levels (r=0.581, P=0.023) in GroupB in12 hours, The percentages of pancreatic acini oncosis in Group C were decline compared with GroupB at 6 hours,12 hours and 24 hours(P=0.040,P=0.017, P=044). The percentages of pancreatic acini oncosis in GroupDalso decline compared with GroupC at 6 hours,12 hours and 24 hours(P=0.033,P=0.004, P=0.027)Conclusion:HMGB1 can induce oncosis of pancreatic acini cell with inducing Inflammatory reaction in the rat with SAP. It is necessary for HMGB1 play its own part to activate NF-κB. HMGB1 may induce oncosis by the activation of NF-κB. |
