Objective: To investigate the frequency of Myeloid-derived suppressor cells and its clinical significance in the peripheral blood of esophageal carcinoma patients.Methods: The specimens of 78 esophageal cancer patients and 15 healthy adults were collected in the Department of Cardiothoracic Surgery of the First Affiliated of Soochow University. Make the expression pattern of peripheral blood white cells (PBWC) in HLA-DR-CD14-CD11b+CD33+ as monocyte derived MDSC, and CD14+HLA-DR- multi-core sources MDSC. Flow cytometry was used to detect and analyze the frequency of MDSC in peripheral blood of two groups. Meanwhile, it also examines MDSC level at 30th day after operation in esophageal cancer.Results: 1.When make the PBWC express CD14+HLA-DR- be MDSC, the proportion of MDSC of esophageal cancer patients was significantly higher than healthy groups, the proportion of MDSC ofⅢandⅣperiod esophageal cancer patients was higher than the proportion ofⅠandⅡperiod patients, the peripheral blood of esophageal cancer patients with lymph-node metastasis express much more MDSC than not. The proportion of MDSC of esophageal cancer patients decreased obviously at the 30th day after operation, and was higher than before the operation.2.Among all the groups, no difference was found at the level of HLA-DR-CD14-CD11b+CD33+ MDSC of esophageal carcinoma.Conclusion: 1.The proportion of MDSC in peripheral blood of esophageal cancer patients is significantly higher than healthy people, moreover it is closely related to the progression of tumor and lymph-node metastasis.2.The proportion of MDSC decreased after resection, which suggests that the immune function recovered in a certain extent.3.It provides a new clue for researching the effect of MDSC in esophageal cancer immunopathology.PartⅡThe phenotypical and functional characteristics of Myeloid-derived suppressor cellsObjective: To study the expression of Costimulatory molecules B7 family on the surface of Myeloid-derived suppressor cells in peripheral blood of the patients with esophageal carcinoma, and its functional characteristics.Methods: The specimens of 5 esophageal cancer patients and 3 healthy adults were collected in the Department of Cardiothoracic Surgery of the First Affiliated of Soochow University. Flow cytometry was used to detect and analyze the frequency of B7 family on MDSC in peripheral blood of two groups. Meanwhile, we also examine the suppression to T lymphocyte proliferation of MDSC by Cell Counting Kit-8, and the expression of cytokines in serum by ELISA.Results: 1.The proportion of B7-H1 on MDSC of esophageal cancer patients was significantly higher than healthy groups.2.MDSC characterized by CD14+HLA-DR- phenotype inhibit the activity of T lymphocyte obviously.Conclusion: 1.Intervention of MDSC may provide a new way of esophageal cancer immunotherapy.2.The high expression of B7-H1 on MDSC of esophageal cancer patients maybe the reason induce MDSC-mediated immunosurveillance.
