Objective:To investigate the frequency of myeloid-derived suppressor cells(MDSC) and its clinical significance in the peripheral blood and cancer tumor of cervical carcinoma patients by flow cytometry.Methods:Collect human peripheral blood from 20 healthy people and 17 patients with cervical cancer untreated, and cervix uteri tissue from 15 cervical tissue from bnign gynecologic tumor and 17 cervical cancer tissue. Organize the tissue into a single cell suspension. Flow cytometry analysis the proportion of overall CD45~+HLA-DR-CD33~+CD11b~+MDSC and subpopulation in the tissue cell suspension in 17 patients with cervical cancer and 15 healthy cervical tissues. The proportions of total HLA-DR-CD33~+CD11b~+MDSC in the peripheral blood of 17 patients with cervical cancer pretherapy and 20 healthy people were evaluated. Test and compare the proportions of CD15~+ granulocyte MDSC and CD14~+ monocytes MDSC’s level in the peripheral blood. Analyze the correlation between the proportion of MDSC in patients’ peripheral blood and the value of squamous cell carcinoma antigen(SCCA)in the cervical cancer group.Results:Compared with healthy volunteers, the proportions of circulation MDSC from cervical cancer were higher(P < 0.01). The frequency of MDSC was positively correlated with the frequency of value of squamous cell carcinoma antigen in the cervical cancer group(r=0.7154, P< 0.05). And both healthy people and patients with cervical cancer, the MDSC in peripheral blood is mainly CD15 ~+ granulocyte subgroup. And the proportions of tissue cell suspension CD45~+ cell from cervical cancer were higher, compared with cervical tissue from bnign gynecologic tumor cellsuspension(P<0.05);But in tissue cell suspension CD33~+cell was not found.Conclusions:MDSC may become an index to evaluate the efficacy of the cervical cancer treatment and the prognosis of patients with cervical cancer, to monitor of recurrence of cervical cancer. Granulocyte subgroup MDSC in the peripheral blood of patients with cervical cancer may closely correlate with the development of cervical cancer.
