The radiotherapy sensitization ofβ-elemene has been confirmed by basic research. But its research of radiotherapy sensitization mechanisms is mainly in vitro stage. Radiotherapy sensitization mechanism, is not clearly explained due to its role in many targets. There are many unknown fields for worth further exploration, To examined whether the effects ofβ-elemene combined with radiotherapy is associated with inhibition of angiogenesis.Objective: To current study examined whether the effects ofβ-elemene combined with radiotherapy is associated with inhibition of angiogenesis by establish the human lung adenocarcinoma A549 cell line xenograft tumor in nude mice.Methods: The xenograft tumor model was established with human lung adenocarcinoma A549 cell line through cell suspension inoculation. The growth and shape of the tumor was observed and the growth curves of tumor in the nude mice were drew.The transplanted female mice with 0.8~1.0cm3 tumor were randomly divided into four groups(5 per group): the control group,radiation alone,β-elemene alone group and combined treatment group (radiation afterβ-elemene). RT-PCR was used to detect mRNA expressions of VEGF and Western blot was used to detect protein expressions of VEGF. The expression of VEGF and CD34 in different groups was detected by immunohistochemistry.Results:1.The xenograft tumor models were successfully established and the growth curve was obtained according to tumor size. The volume of the tumor reached 0.8 ~ 1.0cm3 after about 4 ~ 6 weeks. .2.RT-PCR results showed that the level of the VEGF mRNA inβ-elemene alone group is higher, the difference is statistically significant (P <0.05).the level of VEGF mRNA in the radiotherapy group was significantly higher than other groups (P <0.01), while that in the combination group was significantly lower than the other control group (P <0.01).3.Western blot experimental results showed that The expression of VEGF inβ-elemene alone group was slightly lower than that in the control group, but there was no significant difference (P> 0.05).The expression of VEGF in the radiotherapy group was significantly increased (P <0.01), while that in the combination group was significantly decreased (P <0.01).4.Immunohistochemistry staining showed that The expression of VEGF inβ-elemene alone group was slightly lower than that in the control group, but there was no significant difference (P> 0.05).The expression of VEGF in the radiotherapy group was significantly increased (P <0.01), while that in the combination group was significantly decreased (P <0.01).5.Immunohistochemistry staining showed that The expression of CD34 inβ-elemene alone group was slightly lower than that in the control group, but there was no significant difference (P> 0.05).The expression of CD34 in the radiotherapy group was significantly increased (P <0.01), while that in the combination group was significantly decreased (P <0.01).Conclusion:1.The radiosensitizing doses ofβ-elemene has mild inhibitton to VEGF protein. radiation has obvious promotion to VEGF protein expression.β-elemene combined with radiotherapy has significant inhibitton to VEGF protein expression.2.The radiosensitizing doses ofβ-elemene has mild inhibitton toCD34. radiation has obvious promotion to CD34.β-elemene combined with radiotherapy has significant inhibitton to CD34.3.The effects ofβ-elemene combined with radiotherapy is associated with inhibition of angiogenesis to improve tumor radiation sensitivity...
