Effects Of Hypothermia Resuscitation On PPAR-γ And Related Gene Expression In Rats Lung During Hemorrhagic Shock

Objectives:The aim was to investigate the effects of different body temperature resuscitation on PPAR-γ, NF-κB, TNF-αand iNOS gene expression in the rats lung tissue during hemorrhagic shock.Methods:60 adult female Wistar rats were randomly assigned to three groups(n=20): control group(sham), normalthermia group(NR) and hypothermia group(HR). Rats in the control group underwent the surgical procedure but were not bled. Rats in the normalthermia group and hypothermia group were received normalthermia (38-39℃) resuscitation or hypothermia(33-34℃) resuscitation respectively during resuscitation and hourly thereafter. Using a pressure transducer for the measurement of mean artery pressure (MAP). real-time PCR method detects the PPAR-γ, NF-κB, TNF-αand iNOS mRNA expression in the rats lung tissute after surgical operation, hemorrhagic shock 90min, resuscitation 60min and 240min.Results:1.MAP Changes: There were not significantly different among groups 15 min after of the surgical operation (P>0.05).During hemorrhagic shock the two groups MAP were maintained from 40 to 45mmHg. 10 min after resuscitation the MAP was increased to group NR:101±13.2 mmHg and group HR110±15.1 mmHg. Afterthat there was a progressive decline in MAP to approximately group NR:77±17.2 mmHg and group HR:92±13.5 mmHg at the end of the experiment. During the resuscitation the MAP of the rats in the group HR was higher than the group NR at all the time point, but there were not significantly different (P>0.05).2.PPAR-γmRNA expression: There was PPAR-γmRNA expression in the lung of the normal rats. 90min after of the hemorrhagic shock there was a reduction of PPAR-γmRNA expression in lung of the two groups compared with the normal rats. The dates were 0.83±0.09 times and 0.86±0.07 times compared with the normal rats respectively(P>0.05). 60min after of the resuscitation,the PPAR-γmRNA expression in lung of the two groups were 0.61±0.04 times and 1.71±0.51 times compared with the normal rats respectively(P<0.05). 240min after of the resuscitation, the PPAR-γmRNA expression in lung of the two groups were 0.54±0.08 times and 2.83±0.76 times compared with the normal rats respectively(P<0.05)3.TNF-αmRNA expression: There was TNF-αmRNA expression in the lung of the normal rats. 90min after of the hemorrhagic shock there was an increase of TNF-αmRNA expression in lung of the two groups, compared with the normal rats. The dates were 2.51±0.22 times and 2.63±0.36 times compared with the normal rats respectively(P>0.05). 60min after of the resuscitation, the TNF-αmRNA expression in lung of the two groups were 4.12±0.13 times and 3.36±0.19 times compared with the normal rats respectively(P<0.05). 240min after of the resuscitation, the TNF-αmRNA expression in lung of the two groups were 5.87±0.21 times and 4.91±0.26 times compared with the normal rats respectively(P<0.05).4.iNOS mRNA expression: There was iNOS mRNA expression in the lung of the normal rats. 90min after of the hemorrhagic shock there was a increase of iNOS mRNA expression in lung of the two groups, compared with the normal rats. The dates were 1.77±0.44 times and 1.68±0.53 times compared with the normal rats respectively(P>0.05). 60min after of the resuscitation the iNOS mRNA expression in lung of the two groups were 3.43±0.33 times and 2.21±0.26 times compared with the normal rats respectively(P<0.05). 240min after of the resuscitation, the iNOS mRNA expression in lung of the two groups were 4.86±0.62 times and 2.98±0.27 times compared with the normal rats respectively(P<0.05).5.NF-кB mRNA expression: There was NF-кB mRNA expression in the lung of the normal rats. 90min after of the hemorrhagic shock there was a increase of NF-кB mRNA expression in lung of the two groups, compared with the normal rats. The dates were 1.63±0.56 times and 1.69±0.49 times compared with the normal rats respectively(P>0.05).after 60min of the resuscitation,the NF-кB mRNA expression in lung of the two groups were 3.12±0.31 times and 2.13±0.19 times compared with the normal rats respectively(P<0.05). after 240 min of the resuscitation, the NF-кB mRNA expression in lung of the two groups were 4.33±0.43 time and 2.68±0.38 times compared with the normal rats respectively(P<0.05).6. Lung Tissue Histological Evaluation 240min after of the resuscitation, NR group rats had a significant alteration of lung architecture as demonstrated by interstitial swelling and inflammatory cell infiltration when compared with sham rats. In NR group hypothermia resuscitation attenuated the changes in lung interstitial swelling and inflammatory cell infiltration compare with NR group.Conclusions:1.Hypothermia resuscitation after hemorrhagic shock can raise the ppar-gamma gene expression and inhibit NF-кB, iNOS, TNF-αgene expression in the rats lung tissue.2.Hypothermia resuscitation after hemorrhagic shock can ameliorate inflammatory response in rats lung tissue.
