The ability of a tumor cell to undergo invasion and metastasis may explain why the cancer means high lethality. Cell migration plays a central role in a wide variety of process, such as embryogenesis, inflammatory response, wound healing, and which is also the basic of invasion and metastasis. To be a adhesive molecμlar, integrin can link the cytoskeleton and ECM throμgh its little cytoplasmic domain and ectodomain to anchorage the cell to the ECM. Additionally, integrin can integrate the signals inside and outside of the cell to regμlate the migration. So, integrin is crucial for the migration process. C-Abl kinase is a non-receptor which can phosphorate many protein to activate them, then lead to F-actin rearrangement or gene expression. Therefore, it participates many physiological processes such as cell apoptosis, differentiation, senescence. Some researches also report that c-Abl may regμlate cell migration.In this articles, with the help of transwell experiments we specμlate that c-Abl kinase may participateαvβ3 integrin mediated migration of A375 cells. According to our Immunofluorescence microscopy and co-immunoprecipitation data, we demonstrate that in resting A375 cells, c-Abl kinase andαvβ3 integrin exists in a same protein complex. Then we construct four GST-linked truncated mutations of c-Abl which are the essential structures for protein-protein interaction. GST-pμll down assay announce that c-Abl interacts withαvβ3 integrin complex via its SH3 domain.
