| Objective: Studying the expression of integrin αvβ3 and CD44 in lung cancer and the bone metastasis tissue by immunohistochemistry, and analysis the correlation between them. Then to explore the relationship of integrin αvβ3 and CD44 with the occurrence, metastasis, tumor size and histological grade of lung cancer. In order to clear the correlation of integrin αvβ3 and CD44 expression with the clinical prognosis.Methods: Choose 55 cases of lung cancer paraffin tissue samples and choose 10 cases of normal lung paraffin tissue samples as a control group, which were taken at Fourth Hospital of Hebei Medical University, Department of Pathology from January 2007 to March 2013. Immunohistochemical staining SP method was adopted to detect the expression of integrin αvβ3 and CD44 in tissue samples of lung cancer and the bone metastases, in which integrin αvβ3 monoclonal antibody of mouse ant-human and CD44 monoclonal antibody of mouse ant-human were applied. Statistical analysis of the relationship of integrin αvβ3 and CD44 with the tumor size, histological grade and metastasis of lung cancer or the bone metastases. Organize and follow-up 55 cases of lung cancer cases, to establish the prognostic factors in lung cancer.Results:1 Integrin αvβ3 was expressed in lung cancer and the bone metastases, the positive rate was 89.1%. The integrin αvβ3 positive rate in lung cancer bone metastasis group was higher than it was in primary lung cancer group. The positive rates were 96.0% and 83.3% respectively. Concluded that integrin αvβ3 expression intensity in the three groups was not all the same. Multiple comparisons between groups found that the expression of integrin αvβ3 were significant differences statistically between normal lung tissue and primary lung cancer or bone metastasis of lung cancer tissues(P<0.001). The expression of integrin αvβ3 was also significant difference statistically between primary lung cancer tissue and lung cancer bone metastasis tissue(P<0.05). In addition, the expression of integrin αvβ3 was no significant difference statistically between diameter of 5cm or less lung cancer tissue and the diameter greater than 5cm one(P>0.05). Analogously, the expression of integrin αvβ3 was no significant difference statistically between male and female(P>0.05). The expression of integrin αvβ3 was no significant difference statistically between patients less than 55 years old and the more than 55 years old one(P>0.05).2 CD44 was expressed in lung cancer and the bone metastases, the positive rate was 85.5%. The CD44 positive rate 92% in lung cancer bone metastasis group was higher than it was 80% in primary lung cancer group. Concluded that CD44 expression intensity in the three groups was not all the same. Multiple comparisons between groups found that the expression of CD44 were significant differences statistically between normal lung tissue and primary lung cancer or bone metastasis of lung cancer tissues(P<0.001). The expression of CD44 was also significant difference statistically between primary lung cancer tissue and lung cancer bone metastasis tissue(P<0.05). In addition, the expression of CD44 was little difference between diameter of 5cm or less lung cancer tissue and the diameter greater than 5cm one, and examined by Kruskal-Wallis rank sum test found no significant difference statistically(P>0.05). Analogously, the expression of CD44 was no significant difference statistically between male and female(P>0.05). The expression of CD44 was no significant difference statistically between patients less than 55 years old and the more than 55 years old one(P>0.05).3 The positive rate of integrin αvβ3 in well, moderately and poorly differentiated lung cancer groups were 87.5%, 88.9% and 100.0% respectively. The three groups were examined(P<0.05). So there is a difference between the histological grades of lung cancer. Multiple comparisons between groups found that there was no significant differences statistically between well and moderately differentiated groups, and the same to moderately and poorly differentiated groups(P>0.05). In other respects, the expression of integrin αvβ3 were significant differences statistically between well and poorly differentiated lung cancer groups(P<0.05).4 The positive rate of CD44 in well, moderately and poorly differentiated lung cancer groups were 68.8%, 83.3% and 100.0% respectively. The three groups were examined by Kruskal-Wallis rank sum test(P<0.05). So there is a difference expression of CD44 between the histological grades of lung cancer. Multiple comparisons between groups found that there was no significant differences statistically between well and moderately differentiated groups(P>0.05). There was significant differences statistically between moderately and poorly differentiated groups, as well as well and poorly differentiated groups(P<0.001).5 Spearman correlation analysis showed: the expression of integrin αvβ3 and CD44 protein were correlated positively in lung cancer tissues(r=0.787, P <0.001).Conclusions:1 The expression intensity of integrin αvβ3 and CD44 in lung cancer was significantly higher than in normal lung tissue.2 The expression intensity of integrin αvβ3 and CD44 in bone metastasis tissue of lung cancer was significantly higher than primary lung cancer.3 The expression of integrin αvβ3 and CD44 showed a gradual upward trend as the tissue differentiation reduced of lung cancer.4 Integrin αvβ3 and CD44 are likely to be able to promote bone metastasis of lung cancer, and is closely related to the degree of malignancy and prognosis of tumors. There is a certain value to judge the prognosis of lung cancer. |
