Objective:To investigate the expression and the relation of transcription factor FoxO3a and bcr-abl fusion gene in CML patients at diagnosis and patients treated with Glivec as well as its clinical significance.Methods:38 patients with various periods CML (28 patients at diagnosis,10 patients treated with Glivec) were enrolled in the study,10 patients with nonhematopathy were used as control subjects.Using SYBR Green I, real-time PCR methods for the detection of Fox03a, bcr-abl and GAPDH. The difference of those genes expression between the groups was analyzed by AACt relative quantity means, in which GAPDH gene was used as control.Results:The expression of bcr-abl mRNA in patients with CML-CP was 7.46 times higher than that of the patients treated with Glivec, the expression of bcr-abl mRNA in patients with CML-AP was 4.17 times higher than that of the patients with CML-CP, the expression of bcr-abl mRNA in patients with CML-AP was 31.11 times higher than that of the patients treated with Glivec,the differences among three groups have apparence significance (χ2=4.251-5.736, P<0.05). However, the expression of FoxO3a mRNA in CML-CP, CML-AP and Glivec treatment groups occupied for 3.82%,18.83%,13.21% of the control group, respectively. The differences among all groups have apparence significance (x2=12.99,q=5.73,13.73,8.00,p<0.01) Low-expression of FoxO3a were closely correlated with over-expression of bcr-abl fusion gene (r=-0.874, p<0.01)Conclusions:The high expression of bcr-abl mRNA was increased in patients with different periods CML; while the expression of CML-BP was significantly higher than CML-CP; The expression of bcr-abl mRNA can be reduced by Gleevec;②The expression of FoxO3a was decreased in patients with CML, while Gleevec can increase the expression of FoxO3a.③The expression of FoxO3a and bcr-abl gene correlated negatively.
