Citrus aurantium L. var. amara Engl (C. aurantium L. var amara), a member of genus Citrus (Rutaceae), is the variant of Citrus aurantium L, which contains the similar chemical composition and pharmacological role with dry unripe (or) ripe fruit of Citrus aurantium L. Fruit of C. aurantium L. var amara have been used in traditional medicines for the effectiveness of digestant and expectorant. Its flower is rich in volatile oil, flavonoids, volatile oils, limonin, steroids, thiophenes, etc.. At present, there are few studies on the flower of C. aurantium L. var amara, especially on its pharmacological effects of chemical composition. Study on bioactivity ofβSitosterol(βS), Limonexic acid (LA), and 5 Hydroxy 6,7,3',4' tetramethoxyflavone (HTF) from Citrus aurantium L. var. amara Engl provides theoretical references for the application of flower of C. aurantium L. var amara in more areas.Reducing capacity, inhibition of lipid peroxidation, scavenging of oxygen radicals and the protection effect of damage of H2O2 induced PC12 cell were used to evaluate the in vitro antioxidant capacity of three components. The results indicated that HTF and LA have remarkable inhibition effects on reducing of cell viability rate and T SOD content and rising of MDA content, especially the inhibition rate of reducing of T SOD reached 65.1% for HTF with the concentration of 100 g/mL, by contrast the inhibition was 86.05% for LA. The research of antioxidant activity laid a foundation for further study.The activity of inhibiting tumor and cancer cell proliferation were researched. The inhibition rate of HTF reached 73.0% and 71.0% for B16 and Hela at the concentration of 100 g/mL, The impact of it were better than 5 Fluorouracil (5 F), moreover the impact of LA ranked only second to 5 F.βS inhibited 36.4% proliferation of Hela cell at the concentration of 200 g/mL. In addition, HTF, 5 F, LA andβS indicated an inhibition of 88.2%, 58.6%, 50.3% and 34.2% for SMMC 7721 at the concentration of 200 g/mL. For the capability of protecting the CCl4 induced HL 7702 cell, HTF and LA had effective power to inhibit the rise of GOT, MDA and LDH, their capability increased in a dose dependent manner, and HTF displayed more effective protection effect than Vc as references protection compounds.HTF and LA can effective inhibit the generation of nitricoxide (NO) from LPS activated mouse macrophages (RAW264.7). In addition, they had the capability of inhibiting HBsAg and HBeAg from 2.2.15 cell, butβS had no these capability. The antibacterial research demonstrated that three components had no antibacterial activity.
