Objective:To observe the renal protective effects of Ixeris Sonchifolia Hance onrhabdomyolysis-induced acute renal failure(ARF) in rats.Methods:(1)The animal model of rhabdomyolysis-induced acute renal failurewas estblished by injecting glycerol in hind leg muscle.After24h-waterdeprivation,78SD rats,weighing200±20g,were randomly divided into three groups:normal control group,ARF group, Ixeris Sonchifolia Hance-pretreatment group,with6rets in normal control group,36rats in ARF group,36rats in Ixeris SonchifoliaHance-pretreatment group. Six sub-groups were set up for6h,1d,4d,7d,10d,14d afterglycerol injection in ARF group and Ixeris Sonchifolia Hance-pretreatmentgroup,with6rats in each sub-group.The rats of normal control group received10ml/Kg of0.9%Normal Saline by intramuscular injection.The rats of ARF groupreceived10ml/Kg of50%glycerol by intramuscular injection. The rats of IxerisSonchifolia Hance-pretreatment group received20ml/Kg of Ixeris Sonchifolia Hanceby intraperitoneal injection15minutes before received10ml/Kg of50%glycerol byintramuscular injection(.2)Vena cava sampling was taked out in all cats,and the levelof serum creatinine,urea nitrogen and creatine kinase were determined by thebiochemical autoanalyzer(.3)The kidney tissue was fixed with the4%formaldehydebuffer,and embedded with paraffin.Pathological changes of acute tubular necrosiswere observed by HE staining.(4)ATN scorings were done blindly to measure thelevel of renal tubular damage.(5)The expression of Bcl-2,Bax in renal tissue wereobserved by Immunohistochemical staining(SP methods).Results:(1) The6th hours:The serum creatinine,urea nitrogen,creatinekinase,ATN scorings,the expression of Bcl-2and Bax were74.44±7.03umol/L,12.19±2.60mmol/L,586.09±61.75U/L,1.65±0.58,22.54±2.04%and22.65±2.64%respectively in ARF group,they were66.80±4.11umol/L,9.24±1.76mmol/L,440.37±48.33U/L,1.16±0.41,27.59±3.43%and18.12±2.63%respectively in IxerisSonchifolia Hance-pretreatment group.Comparing with the normal controlgroup(They were52.88±1.99umol/L,6.12±0.86mmol/L,179.82±15.06U/L,0.00±0.00, 31.27±3.78%and13.37±2.41%respectively.),SD rats had higher blood serumcreatinine,urea nitrogen,creatine kinase,ATN scorings and the expression of Bax inARF group and Ixeris Sonchifolia Hance-pretreatment group,and the difference isstatistically significant(P<0.05).SD rats had lower the expression of Bcl-2in ARFgroup and Ixeris Sonchifolia Hance-pretreatment group, and the difference isstatistically significant(P<0.05). ATN scorings was negatively correlated with theexpression of Bcl-2(r=-0.838,P<0.05)and positively correlated with expression ofBax(r=0.813,P<0.05).Comparing with the ARF group,SD rats had lower bloodserum creatinine,urea nitrogen,creatine kinase,ATN scorings and the expression ofBax,higher expression of Bcl-2in Ixeris Sonchifolia Hance-pretreatment group,and the difference is statistically significant(P<0.05).(2)The1st day: The serum creatinine,urea nitrogen,creatine kinase,ATNscorings,the expression of Bcl-2and Bax were280.26±19.04umol/L,24.28±7.11mmol/L,816.17±110.34U/L,3.44±0.56,19.39±2.93%and36.34±4.01%respecti-vely in ARF group,they were209.30±10.61umol/L,16.84±3.83mmol/L,659.52±75.15U/L,3.17±0.36,33.87±3.55%and30.52±3.31%respectively in IxerisSonchifolia Hance-pretreatment group. Comparing with the normal controlgroup(They were2.88±1.99umol/L,6.12±0.86mmol/L,179.82±15.06U/L,0.00±0.00,31.27±3.78%and13.37±2.41%respectively.),SD rats had higher blood serumcreatinine,urea nitrogen,creatine kinase,ATN scorings,the expression of Bax in ARFgroup and Ixeris Sonchifolia Hance-pretreatment group, and the difference isstatistically significant(P<0.05). ATN scorings was negatively correlated with theexpression of Bcl-2(r=-0.854,P<0.01)and positively correlated with expression ofBax(r=0.820,P<0.05).Comparing with the normal control group,SD rats had lowerthe expression of Bcl-2in ARF group and had higher in Ixeris Sonchifolia Hance-pretreatment group, and the difference is statistically significant(P<0.05).Comparingwith the ARF group,SD rats had lower blood serum creatinine,urea nitrogen,creatinekinase,ATN scorings,the expression of Bax,higher expression of Bcl-2in IxerisSonchifolia Hance-pretreatment group, and the difference is statistically significant(P<0.05).(3)The4th day: The serum creatinine,urea nitrogen,creatine kinase,ATN scorings,the expression of Bcl-2and Bax were214.43±26.96umol/L,45.05±7.17mmol/L,730.07±78.64U/L,3.05±0.38,20.22±2.03%,30.91±3.44%respectively inARF group.They were171.38±20.11umol/L,33.82±6.03mmol/L,604.85±41.38U/L,2.83±0.57,36.52±4.08%,26.71±3.25%respectively in Ixeris Sonchifolia Hance-pretreatment group. Comparing with the normal control group(They were52.88±1.99umol/L,6.12±0.86mmol/L,179.82±15.06U/L,0.00±0.00,31.27±3.78%and13.37±2.41%respectively.),SD rats had higher blood serum creatinine,ureanitrogen,creatine kinase,ATN scorings and the expression of Bax in ARF group andIxeris Sonchifolia Hance-pretreatment group, and the difference is statisticallysignificant(P<0.05). ATN scorings was negatively correlated with the expression ofBcl-2(r=-0.811,P<0.05)and positively correlated with expression of Bax(r=0.841,P<0.01).Comparing with the normal control group,SD rats had lower the expressionof Bcl-2in ARF group and higher in Ixeris Sonchifolia Hance-pretreatment group,and the difference is statistically significant(P<0.05).Comparing with the ARFgroup,SD rats had lower blood serum creatinine,urea nitrogen,creatine kinase,ATNscorings,the expression of Bax,higher expression of Bcl-2in Ixeris SonchifoliaHance-pretreatment group, and the difference is statistically significant(P<0.05).(4)The7th day: The serum creatinine,urea nitrogen,creatine kinase,ATNscorings,the expression of Bcl-2and Bax were128.76±21.26umol/L,19.55±5.25mmol/L,633.15±63.01U/L,2.60±0.32,23.16±3.87%,27.39±4.66%respectively inARF group,they were104.93±10.87umol/L,13.63±3.57mmol/L,462.51±41.99U/L,2.00±0.33,34.66±3.67%,23.48±3.62%respectively in Ixeris Sonchifolia Hance-pretreatment group. Comparing with the normal control group(They were52.88±1.99umol/L,6.12±0.86mmol/L,179.82±15.06U/L,0.00±0.00,31.27±3.78%and13.37±2.41%respectively.),SD rats had higher blood serum creatinine,ureanitrogen,creatine kinase,ATN scorings and the expression of Bax in ARF group andIxeris Sonchifolia Hance-pretreatment group,and the difference is statisticallysignificant(P<0.05). ATN scorings was negatively correlated with the expression ofBcl-2(r=-0.812,P<0.05)and positively correlated with expression of Bax(r=0.797,P<0.05).Comparing with the normal control group,SD rats had lower the expressionof Bcl-2in ARF group and higher in Ixeris Sonchifolia Hance-pretreatment group,and the difference is statistically significant(P<0.05).Comparing with the ARFgroup,SD rats had lower blood serum creatinine,urea nitrogen,creatine kinase,ATNscorings,the expression of Bax,and higher expression of Bcl-2in Ixeris SonchifoliaHance-pretreatment group, and the difference is statistically significant(P<0.05).(6)The14th day: The serum creatinine,urea nitrogen,creatine kinase,ATNscorings,the expression of Bcl-2and Bax were55.37±3.27umol/L,6.85±1.34mmol/L,327.54±26.81U/L,1.34±0.40,26.42±3.57%,17.42±2.59%respectively in ARF group.Comparing with the normal control group(They were52.88±1.99umol/L,6.12±0.86mmol/L,179.82±15.06U/L,0.00±0.00,31.27±3.78%and13.37±2.41%respectively.),SD rats had higher creatine kinase,ATN scorings and the expression ofBax,lower expression of Bcl-2,and the difference is statistically significant(P<0.05),they were53.68±2.06umol/L,6.25±0.78mmol/L,183.57±17.62U/L,0.75±0.42,32.87±3.27%,13.56±2.17%respectively in Ixeris Sonchifolia Hance-pretreatment group. ATN scorings was negatively correlated with the expression ofBcl-2(r=-0.783,P<0.05)and positively correlated with expression of Bax(r=0.778,P<0.05).Comparing with the normal control group,SD rats had higher ATN scorings,and the difference is statistically significant(P<0.05). Comparing with the ARFgroup,SD rats had lower ATN scorings,the expression of Bax,higher expression ofBcl-2in Ixeris Sonchifolia Hance-pretreatment group,and the difference isstatistically significant(P<0.05).Conclusions:The expression of Bcl-2and Bax were correlated with pathologychange of renal tissue in rhabdomyolysis-induced acute renal failure.IxerisSonchifolia Hance can reduce kidney injury,improve the renal function,upregulate theexpression of bcl-2ang downregulate the expression of bax in renal tissue of rats withrhabdomyolysis-induced acute renal failure.
