【Objective】Relying on our groups previous experiment: Affymetrix-Rat230cDNA gene chip technology screened TDVT (Traumatic Deep Venous Thrombosis) rats vein endothelial tissue differrential lyexpressed gene, on theBasis of inspection Ncibi Gene and Gene Card database(2012) to determine and platelet activation and endothelial cell adhesion is closely related to members of the selectin family, P-selectin (levels of CD62P) as a further study. Establish the standard, using of RT-PCR technology testing expression of CD62P in thrombosis patients and healthy people' s blood cells which from molecular level to discuss CD62P's molecular mechanism in transcription of deep vein thrombosis process, platelet active participation and cellular adhesive and CD62P whether the specific molecular markers of clinical prediction and early diagnosis of DVT to provide a theoretical reference.[Materials and methods]First:Determine the experimental study to collect the blood samples of each experimental group by RT-PCR to detect CD62P gene expression in white blood cell and platelet transcription1,Reference2012edition of the American College of Chest Physicians Association of antithrombotic thromboprophylaxis clinical practice guidelines-the diagnosis of deep vein thrombosis DVT diagnostic criteria for the development of this research into high-risk groups according to the following risk factors to assess:of thrombosis personal history or family history of prolonged bed rest or braking, age, obesity; acute medical factors:respiratory failure, acute stroke, acute heart failure, acute myocardial infarction, acute infectious diseases, acute rheumatic diseases; medical foundation and chronic factors:chronic effort failure, cancer, paralysis, chronic lung disease, diabetes, obesity, collagen vascular disease and thrombophilia, and inflammatory bowel disease, nephrotic syndrome, metabolic syndrome and other diseases. In addition, mechanical ventilation, central venous catheter, a permanent pacemaker implantation, oral contraceptives, hormone replacement therapy,6weeks of pregnancy and postpartum. Surgical risk factors include general surgery (guitar, rectal cancer), orthopedic surgery, vascular surgery (involving the lower extremity venous surgery), gynecological surgery (childbirth, pelvic tumors), neurosurgery (limb movement disorder and intracranial hypertension. dehydration therapy); orthopedic surgery including total hip and knee arthroplasty, femoral neck fracture and intertrochanteric fracture, femoral shaft fracture, pelvic fracture, spine fracture with spinal cord injury, operative time was45min, intraoperative tourniquet. Assessing risk factors, clinical manifestations (lower extremity pain, swelling, Homans sign, The Neuhof sign) and color Doppler ultrasonography diagnosis. Group A compared with group B, P <0.05; group A compared with group C, P <0.05; group C compared with group B, P<0.05.【Conclusion】Human white blood cells in each experimental group, platelet CD62P gene transcriptional expression by RT-PCR results showed that the group and trauma:thrombosis, blood clots do not form a group with healthy people, compared with CD62P showing significantly upregulated trend in the level of gene transcription, deepvein thrombosis group expression is greater than in patients after trauma after major orthopedic surgery. Accordingly, we initially speculated that the gene may be formed in the DVT and the stress response plays an important role in the closely related with thrombosis.
