Background and Objective:14-3-3proteins are a group of highly conserved acidic protein family which arepresent in all eukaryotes, the molecular weight of28-33KD., There are seven kinds ofsubtypes(β, γ, ε, η, σ, θ, ζ) in mammals.14-3-3proteins interact with a variety ofcellular proteins by phosphorylation and phosphorylation.These cellular proteinsincluding transcription factors, biosynthetic enzymes, cytoskeletal proteins, signalingmolecules, apoptosis factors and tumor suppressors., Many studies have demonstratedthat14-3-3may also play an important role in cancer development and progression,but few reports about gastric carcinoma.In the present study, the expression of14-3-3isoforms mRNA and proteins were observed in human gastric carcinoma and inpaired normal adjacent gastric tissues,and explored the correlation and significancebetween14-3-3proteins expression level and human gastric carcinoma.Methods:Ten human gastric carcinoma samples and ten paired normal adjacent gastrictissues obtained from the gastrectomy.The levels of all seven14-3-3isoforms mRNAwere examined by RT-PCR,and after statistical analysis, the protein expression wereidentified by Western Blotting to those isoforms whose mRNA expression aredifferent.At the same time the isoforms were identified by Immunohistochemistry toobserve the cellular localizationResults:A basal expression of all the seven14-3-3isoforms mRNA were detected byRT-PCR both in the gastric cancer tissues and paired normal adjacent gastric tissues.However the expression level of14-3-3θ and ζ mRNA were significantly discreasedin gastric carcinoma tissues comparing with the paired normal adjacent gastric tissues(P<0.05).The protein expression level of14-3-3θ and ζ were also significantlydiscreased in gastric carcinoma tissues comparing with the paired normal adjacentgastric tissues by Western Blotting and Immunohistochemistry(P<0.05). Conclusions:1. There was a basal expression of all the seven14-3-3isoforms mRNA both inhuman gastric carcinoma and paired normal adjacent gastric tissues, which may berequired to maintain normal gastric function.2. The mRNA and protein expression of14-3-3θ and ζ in gastric carcinomatissues were down-regulation,which suggested those two isoforms of14-3-3proteinsmay be involved in the development of the human gastric carcinoma.
