Chinese mosla Herb belongs to mosla plants of labiatae, which is one the most important erect herbs of herbal medicine. Ti is a precondition of improving additional value to research the methods of extraction and separation, and pharmacological activities of flavonoids in Chinese mosla Herb. The extraction and separation of flavonoids in Chinese mosla Herb has rare reported in China, and the pharmacological activities has nerver reported. The extraction and antioxidant activities of flavonoids in Chinese mosla Herb were studied, and the antioxidant activities of flavonoids in Chinese mosla Herb were compared with TBHQ and tea polyphenol, which provides theoretical guidance on exploitation and utilization of flavonoids in Chinese mosla Herb. Many important conclusions were obtained as follows:(1) The total flavone in Chinese Mosla Herb was extracted by means of alcohol heating extraction. The best extraction method was selected by single factor examination. Based on this single factor experiment, the effects of alcohol concentration, temperature and the ratio of solid-fluid on the extraction were discussed by Response Surface Analysis(RSA), and the best extraction method of these three factors with reasonable values was modified. The results showed that best extraction parameters of total flavone were as follows:extracting time1h,77.6%ethanol as extractant, extraction temperature73.5℃, ratio of solid-fluid1:23.2(m: V), Under this condition,the extraction rate was5.36%.(2)To establish a technical process for the separation and purification of total flavones from Chinese Mosla Herb, the total flavone in Chinese Mosla Herb were separated and purified by resin. The purity and yield of flavonoids were examined by static and dynamic experiments. The results showed D301type resin was the best resin for separating and purificating flavonoids from Chinese Mosla Herb. The static saturated adsorption capacity was104.72mg·g-l, the eluant was70%alcohol, the pH value of sample was5.0. the temperature was30℃, Static adsorption reached a balance in about120min, The dynamic adsorption-Dissolve experiments indicated the optimum conditions were as follows:the volume of the water was4BV, the volume of70%of ethanol was4BV. The recovery and the purity of flavonoids were both more than80%.(3)The experiment of antioxidative activity of flavonoids in Chinese Mosla Herb showed that its reduction rate enhanced as the increase of the of flavonoids content. In the DPPH free radical scavenging experiment, the capability of scavenging DPPH radicals with tea polyphenol was slightly higher than TBHQ and flavonoids. While the abilities of inhibiting linoleic acid peroxidation with tea polyphenol, TBHQ and flavonoids were almost identical their anti-oxidization effects were tea polyphenol> flavonoids in Chinese Mosla Herb> TBHQ. Above research result indicates that the total flavonoids in Chinese mosla Herb was5.36%, D301macroporous adsorptive resins could isolation and purification flavonoids in Chinese mosla Herb efficiently, and the flavonoids had good antioxidant activities, the Chinese mosla Herb can be planted extensively and has great value for deeply utilization.
