| This paper took Tabersonine as experimental subject and mainly engaged in Qualitative and Quantitative Analysis and Study on Extraction and Separation of Tabersonine in Voacango Africana Stapf.. in brief the results as follows:1. Established the operational condition of TLC of Tabersonine, which could be used for qualitative analysis, and the developer was:hexane:ethyl acetate=4:1(V:V).2. The content of Tabersonine in Voacango Africana Stapf was determined by ultraviolet spectrophotometry, which included:the UV examination wave-length is275nm and the UV examination temperature is20℃. Take the absorbency to consistency and get the regression equation Y=148.76X-5.0549(r=0.9997). The result had shown that it has good linear relationship in the range of100ug-mL-1~200μg-mL-1.3. The content of Tabersonine in Voacango Africana Stapf was determined by TLCS.The developer was:hexane:ethyl acetate=4:1(V:V). Used the method of single wavelength zigzag scanning, λs=275nm. Take quality to peak area and get the regression equation: Y=79.953x+1191.8(R2=0.9989). The calibration curve for tabersonine showed good linearity in the ranges of2-10μ.g.4. To establish HPLC method for the determination of tabersonine in Voacanga africana Stapf. The analytes were separated on a Inertsil C18(4.6mmx250mm,5μm) and detected at275nm. The mobile phases were acetonitrile-O.1mol/Lammonium carbonate solution (7:3, v/v). The velocity of flow was0.7mL·min-1. Take consistency to peak area and get the regression equation:Y=15749X—1092, r=0.9995. The calibration curve for tabersonine showed good linearity in the ranges of0.02-0.2mg/mL.5. The optimum extraction condition was investigated by orthogonal design and the extraction quantity of tabersonine was regarded as the investigated index to optimize the extraction technology for tabersonine of Voacanga africana Stapf. The optimal extraction technology was as follows:12-folds of hexane, at temperature of70℃for3times,2hours each time.6. To study on separation and purification of tabersonine in Voacanga africana by multiple extraction method and macroporous adsorption resin. The optimum parameters of the purification process were established as:extacting the sample liquid three times with2%HC1(3:8, v/v), making the extraction liquid through D101macroporous resin(l:15), using80%ethanol as the desorption solvent and extracting it with hexane after alkalization. The result shows that the average productivity of tabersonine is1.543%, the contant is91.15%, and the conversion is71.31%.7. Studied on the content of tabersonine in Voacango Africana Stapf from different harvest time. The result showed that there were more content of tabersonine in Voacango Africana Stapf collected by mature period. Therefore, it is best to take Voacango Africana Stapf of mature period as material to get tabersonine.8. Established the method and technological process of Tabersonine in Voacango Africana Stapf. |
