Study On Separation And Antioxidation Of Constituents From Mango(Mangifera India L)Leaves

The test used mango(Mangifera India L)leaves as material,mostly researched the extraction method of ultrasonic wave,microwave and hot reflux.The effect of these methods on the contents of flavonoid and polyphenol in Jinhuang mango leaves,perennial honey-mango leaves,Aiwen mango leaves,Tainong mango leaves and the antioxidant activity were studied.On the other hand,crude extracts from the strong antioxidant activity mango leaves which were Jinhuang mango leaves and perennial honey-mango leaves were purified by silica gel column chromatography and TLC and the purification parameters were determined by experiment.Finally,the antioxidant activity of the mango leaves was studied by HPLC.The main results showed:1. The contents of flavonoid and polyphenol in the four varieties of mango leaves by ultrasonic extraction method in the three extraction methods were the most abundant.The respective flavonoid contents of Jinhuang mango leaves,perennial honey-mango leaves,Aiwen mango leaves,Tainong mango leaves was:(52.27±1.18)mg/mL;(36.53±0.78)mg/mL;(13.10±0.45)mg/mL; (8.21±0.16)mg/mL. The respective polyphenol contents of Jinhuang mango leaves,perennial honey-mango leaves,Aiwen mango leaves,Tainong mango leaves was (18.78±0.24) mg/mL;(14.58±0.11) mg/mL;(9.24±0.43) mg/mL;(6.71±0.057)mg/mL.2. The scavenging effect on the hydroxylic free radical(·OH)and the DPPH radical of Jinhuang mango leaves was the strongest in the four varieties,the antioxidation activity of the total flavonoid and the total polyphenol in the extracts was in the order of Jinhuang mango leaves> perennial honey-mango leaves> Aiwen mango leaves> Tainong mango leaves.And the mango leaves had better antioxidation activity than Vc.3.To analyze the effect of the three extraction methods on antioxidation activity in the mango leaves extracts,the IC50value of flavonoid on-OH and DPPH was evaluated.The effect of the antioxidation activity in the mango leaves extracts by ultrasonic extraction method was weaker than the other methods.The respective IC50value of Jinhuang mango,perennial honey mango,Aiwen mango,Tainong mango on·OH was (0.85±0.01)ug/mL,(0.92±0.01) ug/mL,(1.29±0.04)ug/mL,(1.89±0.07)ug/mL. The respective IC50value of Jin-huang mango,perennial honey-mango,Aiwen mango,Tainong mango on DPPH was (3.84±0.06)ug/mL,(6.99±0.06)ug/mL,(9.42±0.11)ug/mL,(12.24±0.06) ug/mL. The effect of the antioxidation activity in the mango leaves extracts by reflux extraction method was the strongest in the three methods. The respective IC50value of Jinhuang mango,perennial honey-mango,Aiwen mango,Tainong mango on·OH was (0.89±0.028)ug/mL,(1.73±0.04)ug/mL,(1.83±0.04) ug/mL (2.72±0.011)ug/mL.The respective IC50value of Jinhuan g mango,perennial honey-mango,Aiwen mango,Tainong mango on DPPH was (5.25±0.08)ug/mL,(7.99±0.01)ug/mL,(10.67±0.15)ug/mL,(12.79±0.14)ug/mL.4.Throug a detailed analysis of color reaction of eluted fractions from silica gel column chromatography,the main antioxidative components of Jin huang mango leaves and perennial honey-mango leaves were flavonoid,dihydroflavone and flavone with vicinal trihydroxy. To separate the mango leaves extracts by using gel column chromatography with gradient elution consisting of petroleum ether, ethyl acetate, methanol and acetic acid (1:4:0.5:0.5)(1:5:0.5:0.5)(1:6:0.5:0.5)(1:7:0.5:05),the contents of flavonoid in the eluted fraction S6from Jinhuang mango leaves and the eluted fraction J4from perennial honey-mango leaves were the most abundant,the respective value was (22.46±3.42) mg/mLand (10.78±0.19)mg/mL.At the same time,the antioxidantion activity of the two kinds of fraction was the most excellent.As the concentration of the sample was1.0mg/mL,the scavenging rate was98.86%and71.54%, respectively.5.The analysis of antioxidantive component in the Jinhuang mango leaves and perennial honey-mango leaves by HPLC showed:the mostly antioxidantive component in the perennial honey-mango leaves might be mangiferin, the mostly antioxidantive components in Jinhuang mango leaves might be quercetin, kaempferol and mangiferin.
