Objective:Study on traditional Chinese medicine extract hygroscopicity and carbohydrate mechanism relationship. Determination extract the saccharide composition of Yiganning aqueous, using flocculation process and alcohol sedimentation technique of purification methods, study the difference of two purification methods for Yiganning aqueous extract between hygroscopicity and carbohydrate.Methods:select Pharmacopoeia included the preparation of Yiganning aqueous extract as a model drug, which through different methods of separation and purifycation to get total polysaccharides, monosaccharide, oligosaccharide, and then drying, crushing for the measurement of the above sample hygroscopic percentage and critical relative humidity(CRH). Use HPLC-ELSD detection, the gradient elution method for the determination of carbohydrate composition.The total sugar content is determined by phenol-sulfuric acid method comparing colors; The HPLC-ELSD method for synchronously determining water-soluble free sugar in Yiganning liquid water extraction.Results:The HPLC-ELSD method had the advantages of high accuracy, good stability and simple operation.The CRH of Yiganning extract was67.3%. The CRH of total polysaccharide, total monosaccharide and total oligosaccharide was78.4%,62.2%,68.3%respectively. Samples of the critical relative humidity is bigger, the substance of the hygroscopic is smaller. Total monosaccharides mainly contains fructose oligosaccharide, small molecules in free sugar of total oligosaccharides mainly contains fructose, glucose, sucrose. The result, the carbohydrate components i one of the factors of for increasing the hygroscopicity of Yiganning dry extract, and the moisture absorption maximum material is fructose. Through the comparison of two processes between flocculation process and technology of alcohol precipitation, chitosan flocculation process for moisture absorption was significantly lower in alcohol precipitation process, and the process of fructose content, solid content of total sugar content, solids retention rates are better than alcohol precipitation process.Conclusion:Chitosan clarification method can degrade the hygroscopic of Yiganning extract, at the same time to better retain sugar content, and then it is provide experimental basis for the study of moisture proof technology in traditional Chinese medicine extract.
