Investigating the influence and regular pattern of two purification processes for water extract from Traditional Chinese medicine and by using extraction rate of extract, chemical composition contents, extract hygroscopicity and HPLC fingerprint as evaluation index.Methods:The monosaccharides and polysaccharides content is determined by phenol-sulfuric acid method with two purification processes; The protein content is determined by coomassie brilliant blue method with two purification processes;The amino acid content is determined by ninhydrin method with two purification processes; The tannins content is determined by complex legal method with two purification processes; The extract moisture absorption curve and critical relative humidity is determined by constant temperature and humidity and constant temperature wet method with two purification processes; The astragaloside, lobetyolin and Chlorogenic acid is determined by HPLC method with two purification processes; The Codonopsis pilosula and Herba artemisiae of fingerprint is established by HPLC gradient elution method, the content of Codonopsis pilosula and Herba artemisiae of two purification processes is evaluated by the relative contents of them.Results:The monosaccharides, polysaccharides, amino acid contents and extract yield are higher by Chitosan flocculation clarification process from the three herbs water extraction; The tannin content, moisture absorption are lower by Chitosan flocculation clarification process from The three herbs water extraction, and the critical relative humidity higher than alcohol lcohol precipitation process; The protein and the effective components transfer rate from the three herbs is different; Through the analysis of Codonopsis pilosula and Herba artemisiae fingerprint comparison, Chitosan flocculation clarification process was expressed better for reserving the high polarity component from water extract, but it is inverse for the low polarity component.Conclusion:The experimental data could be used to chitosan flocculation clarifying method to reduce concrete moisture absorption mechanism of traditional Chinese medicine providing theoretical support and experimental basis for further research, and the chitosan flocculation clarifying method in the production of TCM and to provide the reference.
