Objective:to study the influence of Chinese herbal medicine water extraction and alcohol precipitation method and water extraction chitosan flocculation clarify method to aqueous extract of Paeoniaeã€Moutan Cortexã€Taraxaci Herba. Comparing the two kinds of purification methods yield of extracts, the main effective composition, main invalid composition of the transfer rate, extracts of moisture absorption change rule, and two of the whole purification methods chemical composition of the change rule.Methods:HPLC method was used to determine the content of herbaceous paeonifloriã€paeonolum and caffeic acid on the extracts by water extraction and alcohol precipitation method and water extraction chitosan flocculation clarify method;Phenol-sulfuric acid method was used to determine the monosaccharide content and polyose content on the tow extracts; Ferrocenylmethyl-substituted indenes three ketone method was used to determine the Amino acid content on the two extracts。 Coomassie brilliant blue method was used to determine the protein content on the two extracts; Using the constant temperature and humidity and constant temperature wet method to calculate the moisture absorption curve, moisture absorption equation, moisteure absorption acceleration, critical relative humidity on the two extracts;Using HPLC gradient elution method established finger-print of Paeoniae, the composition of the relative content and total material apparent relatively apparent content evaluation two purification methods to the influence of the root of herbaceous peony component content.Results:The dry extract yielding rate of chitosan flocculation clarify method was significantly higher than alcohol precipitation method for the three Chinese herbal medicine; Herbaceous deoxyuridine rate of chitosan flocculation clarify method was higher than alcohol precipitation method, but the transfer rate of paeoniflorin of moutan cortex and caffeic acid of tarxaci herba were all low than alcohol precipitation method.Transfer rate of amino acids, protein, simple sugars, total polysaccharides of the medicinal material for different and different.The percentage moisture of Chitosan clarification samples were low than ethanol precipitation samples. Through the analysis of the root of herbaceous peony fingerprint, we find the flocculation to hydrophilic composition is larger than reserves by alcohol precipitation method, and the reserves of medium polarity part no significant difference between the two methods,the flocculation to lipotrepy compowition is less than veserves by alcohol precipitation method.Conclusion:The experimental data provides theory support and experimental basis for a further research the mechanism of chitosan flocculation clarify law reduce moisture absorption.And provides experimental and theoretical basis for the application of pharmaceutical technology by chitosan flocculation clarify method.
