Chitosan is prepared from the chitin by deacetylation in alkaline mediea which is mainly distributed in the shell of shrimp and crab. Because of its good biocompatibility, antimicrobial activity, biodegradability and many unique physiological function, it has been widely used in medicine, food, agriculture, environmental protection and other fields. Traditional Chinese herbal medicine is one of the treasures of our Chinese people, which plays a very important role in preventing and treating disease. The clarification process of Chinese herbal medicine extract is an important part of development and utilization of traditional Chinese herbal medicine. Chitosan has an excellent flocculation property, which can overcome the traditional alcohol precipitation method in the presence of high cost, large amount of labor, loss of functional ingredients and so on. As a kind of natural product and food additive, it is undoubtedly a green technology which conforms to the requirements of environmental protection when it is used to clarify the extracting solution of Chinese traditional medicine. With the retention rate of functional components and light transmittance of the extracting solution as indexes, the optimum condition of clarification process of extracting solution from Rhizoma Chuanxiong, Houttuynia and Flos Sophorae with chitosan was investigated using single factor experiment and orthogonal experiment. At the same time, by monitoring the change of pectin, protein and polysaccharide content in the clarification process of Flos Sophorae extract, and combined with Fourier transform infrared spectrum, the main components of the flocculation was explored. We observed the change of the particles in the extract in the process of clarification by optical microscope, and studied the mechanism of flocculation and removal of impurity by chitosan. The specific contents and findings are as follows:(1)The optimization of clarification process of extracting solution from Rhizoma Chuanxiong with chitosan.The optimum condition of clarification process of extracting solution from Rhizoma Chuanxiong Houttuynia with chitosan was studied by single factor experiment and orthogonal experiment with the retention rate of ligustrazine and the light transmittance of extracting solution as indexes. The optimum conditions of flocculation were as follows: the amount of chitosan was 1.33g/L, the pH value was 4.0 and the clarification temperature was 50℃. Under this condition the retention rate of ligustrazine was 62.70% and the light transmittance was 72.30%.(2)The optimization of clarification process of extracting solution from Houttuynia with chitosan.With the retention rate of flavonoid and the light transmittance of extracting solution as indexes, the optimum condition of clarification process of extracting solution from Houttuynia with chitosan was investigated by single factor experiment and orthogonal experiment. The results as follows: the dosage of chitosan was 0.4g/L, the p H value was 6.0 and the clarification temperature was 60℃. Under this condition the retention rate of flavonoid was 80.00% and the light transmittance was 68.10%.(3)The optimization of clarification process of extracting solution from Flos Sophorae with chitosan.The light transmittance and the retention rate of rutin effects of chitosan on the extracting solution of Flos Sophorae were studied by individual factor experiments. The results show that the optimum technology parameters are chitosan content 0.6g/L, p H6.0 and 60℃. As the above technological conditions, the light transmittance of clarified liquid was up to 93.53% and the retention of rutin was 91.56%.(4)Explored the mechanism of clarification of chitosan on extracting solution herbal medicine.The flocculation of chitosan is comfirmed that it was mainly composed of polysaccharide, protein and pectin, which was characterized by fourier transform infrared spectroscopy(FTIR) and combined with the content changes of pectin, protein and polysaccharide. The changes of particles in clarification process were observed by optical microscope. After adding chitosan, the particles were firstly united together, then flocculated and sedimentated under the effect of adsorption bridge effect.
