Acanthopanax is a traditional Chinese medicine, belonging to the Araliaceae and containing many effective chemical constituents such as flavonoids, saponins, polysaccharides, trace elements and amino acids. It has the effects of anti fatigue, anti-inflammatory, antiviral, anticancer, anti-aging. This essay taked Acanthopanax stems and leaves flavonoids as the research object, studied the extraction and antioxidant activity and laied a good theoretical foundation for the deep development of. Acanthopanax resource(1) Methods the results were showed as following:Organic solvent extraction method:the analysis of orthogonal test optinizaiton which based on the result of single factor experiment showed that the optimum process parameters of acanthopanax stems and leaves total flavone extraction were:the ethanol concentration was65%,the extracting temperature was75℃, the ratio of solid to liquid was1g:30ml and extracting duration was3h, At this condition, the yield rate of acanthopanax stems leaves total flavonoids was0.651%;Enzymatic extraction method::the analysis of orthogonal test optinizaiton which based on the result of single factor experiment showed that the optimum process parameters of acanthopanax stems and leaves total flavone extraction were:the enzyme concentration was0.65mg/ml, enzymatic hydrolysis time was3h, pH of enzyme solution was5.1and enzymatic hydrolysis temperature was45℃, the yield rate of acanthopanax stems leaves total flavonoids was0.597%;Microwave extraction method::the analysis of orthogonal test optinizaiton which based on the result of single factor experiment showed that the optimum process parameters of acanthopanax stems and leaves total flavone extraction were::the ethanol concentration was75%, microwave power was350W, microwave irradiation time was220s and the ratio of sol id to liquid was1g:30ml, the yield rate of acanthopanax stems leaves total flavonoids was0.578%;Ultrasonic wave extraction method:the analysis of orthogonal test optinizaiton which based on the result of single factor experiment showed that the optimum process parameters of acanthopanax stems and leaves total flavone extraction were:the ethanol concentration was65%, ultrasonic time was75min, the ratio of solid to liquid was1g:35ml and the extraction temperature was65℃, the yield rate of acanthopanax stems leaves total flavonoids was0.603%.(2) Through the comparative analysis of four kinds of extraction methods in operation difficulty, target components of the extent of damage, toxic size, acanthopanax stem leaf total flavonoids yield size, organic solvent extraction method was proved to be the best extraction method(3) Using scavenging hydroxy free radical, NO2-ion, DPPH radical and reduction capacity based four kinds of methods to study the antioxidant efficacy of total flavone extracted from Acanthopanax stems and leaves The results showed that the total flavones form acanthopanax stems and leaves could effectively scaveng hydroxyl free radical, NO2-ion, DPPH radical, while it’had reduction ability.when the concentration of flavonoids was0.25mg/ml,the reduction ability was0.75%;when the concentration of flavonoids was0.5mg/ml, the seavenging ability was25.6%; when the concentration volume was2.5ml, the scavenging hydroxyl free radical capacity was62.7%;when the concentration of flavonoids was0.2mg/mL, the scavenging ability on DPPH free radieal capacity was0.62%.
