There are about 26 kinds and 18 subspecies of Acanthopanax Miq. plants belong to Araliaceae in China. The root bark of Acanthopanax gracilistylus W. W. Smith, A. sessiliflorus (Rupr. et Maxim.) Seem, A. henryi (Oliv.) Harms, A. senticosus (Rupr. et Maxim.) Harms, commonly called "Cortex Acanthopanacis", belonging to the liver meridian and kidney channel, are used as usual Chinese traditional herbs, which are warm-natured, acrid flavor, bitter, and. It has the efficacy of clearing wind-damp, nourishing liver and kidney, strengthening the bones and muscles, eliminating hydroncus, antipyretic and analgesic. acankoreanogenin and acankoreoside A, which are extracted from Acanthopanax Miq., have antiphlogistic activity individually and are expected to become a new anti-HMGB1 drug, and the effect of acankoreanogenin is better. For further filtrating the plantorigin and developing a new standard for the quality of plantorigin, we research on the fingerprint of the leaves of A. gracilistylus W. W. Smith and the contents of acankoreanogenin and acankoreoside A in 10 kinds of Acanthopanax Miq. plants by RP-HPLC.In order to find the difference on their constitutes among A. gracilistylus W.W. Smith from different harvest period, the method of HPLC-UV fingerprint was established for quantitative analysis for the first time. The best conditions of HPLC:wavelength is 214 nm, temp is 30℃, flow rate is 1 mL/min, mobile phase is CH3CN-H2O (0-10 min CH3CN 10%,10-60 min CH3CN 10%â†'35%,60~105 min CH3CN 35%â†'65%,105~110min CH3CN 65%). The results shows that the content of Acanthopanax gracilistylus W.W. Smith is different at various harvest time and provides the basis for the best harvest period. It also contributes to the foundation of the new standard for the quality control of A. gracilistylus W.W. Smith.Smashing tissue extraction was primarily used for extracting acankoreanogenin and acankoreoside A from 10 kinds of Acanthopanax Miq. Plants. And the two components were orderly separated by RP-HPLC method for the first time. The best conditions of HPLC for the two components were also established. For acankoreanogenin: wavelength is 214nm, temp is 25℃, flow rate is 1 mL/min, mobile phase is CH3CN:H2O:CH3COOH=600:400:1. For acankoreoside A: wavelength is 218 nm, temp is 40℃, flow rate is 1 mL/min, mobile phase is CH3CN-H2O (0-30 min CH3CN 10%â†'50%,30~40 min CH3CN 50%). The results provide references for the distribution of acankoreanogenin over Acanthopanax Miq. plants.The results of this paper provides important theoretical basis for finding the best harvest period of A.gracilistylus W. W. Smith and are of important significance to find the plant resource of acankoreanogenin. It also has the referred value for further exploiting of bioactivity compon-ents from A. gracilistylus W.W. Smith and other Acanthopanax Miq. plants.
