Acanthopanax is a medicinal and food homogenous plant resource unique to northern China.Its roots,stems and leaves can be used as medicine.Acanthopanax leaves are rich in flavonoids and are natural antioxidants.However,Acanthopanax products that are widely available in the market are relatively scarce and do not attract public attention.In order to promote the use of Acanthopanax senticosus resources,this paper made Acanthopanax fermentatively made into Acanthopanax senticosus tea and studied the effect of its anti-radiation efficacy on human umbilical vein endothelial cells before and after fermentation.The first attempt was made to use Monascus,Aspergillus oryzae,and Candida utilis to conduct solid state fermentation of Acanthopanax senticosus fresh leaves.By measuring changes in flavonoid content during fermentation and changes in the sensory score of tea,the ratio of strains was selected and the fermentation process conditions were optimized..The test results are as follows:The best bacteria addition ratio is Aspergillus oryzae:Monascus:Candida utilis = 1:2:1.15%inoculum,51%moisture content,fermentation temperature 36?,fermentation is obtained for 4.5 days The Acanthopanax Sinensis fermented tea has excellent sensory and flavonoid content,which is 32.37%higher than unfermented Acanthopanax senticosus.Ultrasonic-assisted method was used to study the conditions of water extraction and alcohol flavone extraction from Acanthopanax Senticosus before and after fermentation.The single factor test was conducted from the ethanol volume fraction,material-liquid ratio,extraction temperature,extraction times and extraction time,and the pre-and post-fermentation spines were determined by orthogonal test.The best conditions for the four processes of extracting water and extracting flavonol from Fentiana are as follows:Purify the crude flavonoids of Acanthopanax senticosus and select the best resin type as AB-8 resin,and explore the conditions through static and dynamic purification.Finally,four more pure Acanthopanax flavonoids were obtained.The purity of UAE was 124.55 mg/g,the purity of unfermented UAE was 138.98 mg/g,and the purity of FAE was 337.87 mg/g.The purity of mg/g and FAA was 510.39 mg/g.The purified four Acanthopanax senticosus flavonoids extracts had good antioxidant activity in vitro.The antioxidation ability of the extracts from strong to weak was FAA>UAA>FAE>UAE.Of Four Acanthopanax flavonoids against UV radiation damage in human umbilical vein endothelial cells(Human umbilical vein endothelial cell,HUVEC)impact.The cell viability was measured by MTT assay,malondialdehyde(MDA)content was detected by TBA method,superoxide dismutase(SOD)was detected by WST method,and apoptosis was detected by DNA-Ladder.The results showed that Acanthopanax flavonoids have good anti-radiation efficacy and the preventive effect is better than the repair effect.Protective capacity were:FAA>UAA>FAE>UAE,and dose-dependent relationship with the administration concentration.
