Sapium sebiferum Roxb. for Euphorbiaceae Sapium, Which Orignate from China, has been become one of the most important energy trees in China. It has been distributed in many parts of China. Dabie Mountain in Hubei province is one of the most important palnting areas. In this paper,72 samples of Sapium sebiferum trees have been collected from this area (Dawu, Yingshan, Macheng, Luotian). This study was the first time to analyse the variation and correlation of the Sapium sebiferum trees’raits. Meanwhiles, that analyzed the genetic diversity and the origin of germplasm, Which can be used for evaluation, utilization and protection to Sapium sebiferum in Hubei Province, as well as provide theoretical foundation. The results can be listed as follow:(1) Variability of Sapium sebiferum Roxb in central Dabie Mountains:the Variation quantitative traits in this 4 respectively planting areas were significant. Whereas, the Variation quantitative traits among these 4 planting areas were non significant.(2) The Variability of single-Sapium sebiferum in central Da bieshan regions indicate that:the frequency of trait variations were different among all the quantitative traits of individuals, the length of infructescence was the largest, which reached to 57.70%; the smallest was leaf-indexs, which reached to 4.65%.(3) The principal component analysis to quantitative traits, yield and oil rate of in central Da bieshan regions:the first 6 principal components of cumulative contribution rate was reached to 84.18%. Each principal components were reflected to the fruits of growth, yield, oil rate and the accumulation of photosynthate. The principal components is reasonable and can be reflected characteristic value of Sapium sebiferum Roxb objectively.(4) The Cluster analysis of single-Sapium sebiferum:10 quantitative traits which including quantitative characters such as length of infructescence, fruit number of an infructescence,fruit branch ratio, leaf index, seed number,1000-seed weight have been used for cluster analysis. The results showed that the plant can be generally Clustered into two categories according to fruit cluster.(5) The correlation analysis of quantitative traits with yield and oil rate in central Da bieshan regions:In this study, the correlation analysis showed that the relationship between various traits and yields were significant; the production of individual plants were decided by 3 quantitative characters, such as seed number, length of infructescence and fruit branch ratio. The main production unit of crown and grain weight was decided by 1000-seed weight, length of infructescence and fruit branch ratio. The relationship between main production unit of crown and these three factors was significantly positive correlation. The relationship between oil rate and length of infructescence, the average number of fruit, fruit branch ratio was weak.(6) The system to DNA extraction and purification technology for different Sapium sebiferum tissues:Extraction fluid volume 350ul, phenol-chloroform-isoamylol consumption 350ul, isopropanol consumption 450ul, and then water bath time 50min, RNase A enzyme dosage of TE 10:1 liquid to completely remove the dissolved RNA, in addition to extraction buffer with 1.5%β-mercaptoethanol and 2.5% PVP.(7) The reaction system suits to ISSR-PCR in different Sapium tissues:20μl of PCR reaction mixture included,20ng/ml of DNA template 1μl,2U Taq polymerase 0.2μl, 50mmol·L-1Mgcl 1.5μl, lOmmol·L-1dNTPs 0.2μl,10 mmol·L-1Buffer 2μl,2.0μmol·L-1 Primers 1.5μl, padded with double distilled water to 20μl.(8) Primer screening:useing of 10 primers were four main producing areas of Sapium sebiferum leaves 72 DNA for PCR amplification were obtained 137 clear amplified bands. Which may be statistical in 137 bands, the polymorphic bands was 121, the percentage of polymorphic loci (PPB) was 93.5%, the average polymorphism primer was 88.01%.(9) The analyse to genetic diversity of Sapium sebiferum in central Dabie mountain Hubei Province:the genetic diversity parameters in species level were:PPB=96.82%, AO=1.9682,AE=1.4812,HE=0.2814,HO=0.3889; genetic diversity parameters in the population level were:PPB=85.91%,AO=1.8142, AE=1.3871, HE=0.2188, Ho=0.2554; Therefore, the overall level of genetic parameters of genetic diversity is high.(10) Genetic diversity analysis of four main producing areas:The percentage of polymorphic loci (PPB) in four counties producing areas, Yingshan、Dawu、Luotian、Macheng, were respectively:89.31%,80.96%,68.86%,88.74%. the polymorphism Dawu has highest group (PPB/%=89.31%) among them, followed by Macheng groups (PPB /%=88.74%) and Yingshan group (PPB/%=80.96%), Luotian group (PPB/%= 68.86%) has the lowest polymorphism.(11) The genetic relationship between four main producing areas:the Nei’s genetic identity (Ⅰ) in county and city populations of Sapium sebiferum have been calculated, Luotian has the lower degree of genetic identity than that of in Dawu, Yingshan, Macheng; The genetic distance of four counties can be classified into two large group from UPGMA cluster map. Yingshan and Dawu have the closest genetic relationship and can be aggregated into one group. And Luotian and Macheng can be aggregated into another group according to genetic relationship relationship...
