Microsatellite, generally refers to the genome by a short repeat unit DNA consisting of short tandem repeats. Microsatellite markers, also known as simple tandem repeats (STRs) or simple sequence repeats (SSR), is the eukaryotic genome in the uniform distribution of simple sequence repeat, usually from 2 to 6 nuclear Nucleotide tandem repeats composition. As the number of tandem repeats in the duplication between different individuals within the group showed a high degree of variability and abundance, so microsatellite in animal breeding and genetics, genetic resources and conservation, evaluation, analysis of genetic variation, paternity and court Medicine And so has a very wide range of applications.Microsatellite in all aspects of the application, usually requires multiple microsatellite loci for amplification and detection, amplification of several large time-consuming and laborious, and expensive. If the use of multiplex amplification technology (mutiplex PCR), can significantly improve the detection efficiency and greatly reduce the experimental costs.In this study, we examined 12 microsatellite loci on 144 individuals of swine and 12 microsatellite loci on 123 individuals of goats, screened microsatellite loci suit for test samples groups, and validated them in a sample group, finally we obtained the following results:1) Filter out the right and microsatellite loci were PCR amplified.2) For obtain individual genotypes of the genetic polymorphism analysis with which the two groups of genetic resource information. Pig populations, the average number of alleles was 7.4, power of discrimination was 0.7974, polymorphism information content was 0.6155, the culnulate power of exclusion was 0.9835. Goats population, the average number of alleles was 9.1, power of discrimination was 0.9096, polymorphism information content was 0.7527, the culnulate power of exclusion was 0.9999.3) Multiplex PCR:the pig samples were amplified by a ten- multiplex PCR; goats composite sample was amplified three triple and a single multiplex PCR combination weight; and its possible And accuracy of a preliminary validation. Ten pigs re-multiplex PCR combination of microsatellite loci were SW240, SW951, SW857, S0155, S0386, SW902, S0101, S0225, S0227 and S0355.
