P.×euramericana cv.‘Portugal’ is one kind of fast growing high quality treeswhich has the advantages of fast-growing, straight and beautiful and it has a broadprospect to be embarked. In this dissertation, taking the spring-time neonatal stems andpetioles and leaves as explants, regeneration technology via tissue culture of popularof P.×euramericana cv.‘Portugal’.Major results were as follows:(1) Explants induction differentiation effect is best on the MS with TDZ0.05mg/L+6-BA0.05mg/L+NAA0.05mg/L+sucrose30g/L.The inductiondifferentiation rate of petioles and stems can reach97%and90%, but the inductiondifferentiation rate of leaves is only10%.(2) When clumb buds transform to MS with KT0.5mg/L+GA31.0mg/L+fructose30g/L, the effect of shoots elongation is best. The rate of shoots elongation ofpetioles and stems both can reach90%, and the length of buds can reach to3cm, eachbunch bud has four adventitious buds equally.(3) In vitro plantlets displayed characteristics of short after proliferationculture.The adventitious buds can not take root well when transfer into the medium ofrootage shoud be strong on the MS medium.(4) The best condition of dotage of asepsis plantlets is1/2MS+NAA0.02mg/L+IBA0.05mg/L+IAA0.05mg/L+sucrose30g/L. Rooting rate can reach to94%, withabundant root system and sturdy Plantlet.(5) Those regenerated plants,5-7cm with strong roots and high lignification, weretransferred to greenhouse.And the plants grew well after14days when their new rootsgrew out.The survival rate of transplants could reach90%.
