In order to investigate distribution characteristics of the IgA secreting plasmacells in the Bactrian camel small intestine mucosal. Through the methods ofsedimentation of saturation ammonium sulfate and DEAE-52cellulosechromatography column isolated and purified sIgA from the Bactrian camelcolostrums, the method of isolate and purify sIgA from the Bactrian and sIgAinspection had a further optimized, use the30%and50%saturation of ammoniumsulfate salting-out precipitation and DEAE-52cellulose chromatography columnrapidly purified high concentrations sIgA from the Bactrian camel colostrums.utilizethe PAGE and SDS-PAGE to detect the purification sIgA. After that, using thepurification sIgA prepared rabbit serum against Bactrian camel sIgA, to further ensurethe accuracy of the nest immunohistochemistry staining locate the Bactrian camelsmall intestine mucosal IgA secreting plasma cells, isolated and purified the rabbitserum, and then we got the high concentrations of rabbit anti-Bactrian camel sIgAantibody. Last, through the methods of immunohistochemistry stained and positionedthe Bactrian camel small intestine mucosal IgA secreting plasma cells, use theOLYMPUS BX-UCB/BX61optical microscope and image acquisition system andIBM SPSS Statistics19analyze the related date, finally, determine the distributioncharacteristics of the IgA secreting plasma cells in the Bactrian camel small intestinemucosal.In the process of isolate and purify sIgA from the Bactrian camel colostrums,Combining the SDS-PAGE and PAGE methods identified the structure of Bactriancamel sIgA which contains the J chain, light chain, heavy chain and secretorycomponent, the molecular weight were about15Ku、20Ku、55Ku、75Ku, themolecular weight of Bactrian camel sIgA was about380Ku.Through the immunohistochemistry staining found that, the IgA secreting plasmacells in the Bactrian camel small intestine mucosal were mainly distributed the base ofintestinal villus lamina propria. In the three sections of the small intestine lamina propria, the density of IgA secreting plasma cells showed a trend of decreased afterincreasing, the maximum(9.89±2.47per104μm2) appeared in the middle of jejunum,this result was different from the IgA secreting plasma cells maximum density inpiglet small intestine lamina propria which appeared in the duodenum.Research on the Bactrian camel sIgA molecular weight and the density of IgAsecreting plasma cells in the Bactrian camel small intestine lamina propria was firsttime, further enrich the date about IgA secreting plasma cells in the mucosal immunesubjects.
