| Araceae Amorphophallus are perennial herb plants, and distributed in Indochina, southern Yunnan and Southeast Asia.China is one of the origin center of Amorphophallus. By the long-term natural variability and the cμltivation of domesticated, there are widely variety of Amorphophallus. However, due to have not carried out a survey of varieties, Amorphophallus are seriously homonym and synonyms, and varieties are confusing. DUS testing of new plant varieties, is about the Distinctness, Uniformity and Stability testing of a new varietiesof plant. Also, DUS is a standards and the necessary procedure of new varieties validated. China haven’t developed DUS test guide of new varieties on Amorphophallus. This study, we fieldly surveyed and Concentratly cultivated the Amorphophallus varieties. Then, used morphological traits Classification and ISSR molecμlar markers to formulate DUS test guide on Amorphophallus Konjac varieties. Conclusions are as follows:The frist one, Fieldly surveyed Chongqing, Sichuan, Yunnan, Guizhou, Guangxi, Shaanxi, Hunan, Hubei, Jiangxi, Taiwan Amorphophallus varieties, received 96 Amorphophallus variety materials, including69 region cμltivars,5 validation varieties,6 hybrids varieties 5 wild varieties, and 11 varieties to be identified.The second one, For two years consecuttly observated and statistted on the morphological characters of Amorphophallus varieties, identified 55 characters for numerical taxonomy analysis. Then used PASW Statistics 18 software for the Q-mode cluster, at the grade line L=15, the 96 Amorphophallus varieties material are classlied into 8 categories,And the classification is the same as the Flora of China.The third one, used ISSR molecular marker technology marked 96 varieties of Amorphophallus; used NTSYS the-PC2.10 bio-analysis software for genetic clustering. At the similarity coefficient D=0.753,96 varieties of Amorphophallus can be divided into 9 categories. The classification is the similar as the Q-mode cluster. Verified the number of taxonomic methods and traits selected reasonable. The fourth one, refered to the literature of the UPOV about DUS Test Guide on new varieties of plants. Combined with the the result of Amorphophallus varieties survey, Numerical Taxonomy konjac and ISSR markers, formμlated Amorphophallus new varieties DUS Test Guide. |
