| TFL1(TERMINAL FLOWER1) homologous genes play important roles in maintaining the plantvegetative growth and inflorescence meristem characteristics in several different plant species, the lackof function may lead to early flower and inhibited the normal development of inflorescence. Bambooplants is perennial plant and flowering time is not sure, so the TFL1gene on bamboo floweringmechanism research has the very vital significance. In this study, based on the conservatism of thesequence of the Gramineae plants, we isolated one homologous gene of TFL1from Bambusa oldhamiby rapid amplification of cDNA end (RACE) and RT-PCR, also evaluated its functional characteristics.DNA sequence analysis showed that the entire cDNA length we obtained was833bp and the genomeDNA sequence was1225bp containing a complete open reading frame that encodes a polypeptide of173amino acids, a85bp of5’-and226bp of3’-untranslated fragment. And the amino acid sequenceof the gene forecast, and analyzes the protein secondary structure, and the results show that the proteinhas rich random coil, at the same time, containing beta turn structure, and the (Extended strand.Phylogenetic analysis of TFL1homologous genes based on amino acid sequences revealed that BoTFL1and other TFL1genes of Gramineae plants have a high identity of more than90%.RT-PCR revealed thatBoTFL1have4.5times higher expression in Flower buds than in Vegetative shoot.35S:: BoTFL1transgenic Arabidopsis in T3generations ago all flowered later than wide type, to T3generations afterflowering time compare to the wild type Arabidopsis show no obvious difference, we assume that theBoTFL1may be occurred gene silence in the growth process of Arabidopsis. At the same time flowerbuds and sepals of transgenic Arabidopsis have also had the structure change, show that BoTFL1genemay closely related with the flowering time and flower of the morphological structure of Bambusaoldhami. Reverse mutation experiment also suggests that BoTFL1gene can reply tfl1-13mutantsphenotype of Arabidopsis. |
