The study was based on xylazine combined anesthetic. Inorder to research the effects of xylazine combined anesthetic and its main components on amino acid neurotransmitters in different encephalic regions of goats,65goats were divided into4groups randomly, and injected normal saline, midazolam (14mg/kg), xylazine (12.8mg/kg) and combined anestgetic (1.31mL/kg) respectively. The changes of ethology were observed and the brain tissues were took in different times. The content of amino acid neurotransmitters in different encephalic regions of goats were detected by high performance liquid chromatography (HPLC). According to the trend of changes in different encephalic regions of amino acid neurotransmitters in anesthesia process, it can investigate the possible active site of the xylazine combined anesthetic and its main components, and the possible roles they involved in the anesthesia mechanism.Results:Changes of excitatory amino acids neurotransmitter in different encephalic regions:In xylazine group, Asp content of each encephalic region in goats decreased34%,26%,14%,22%and12%respectively; Glu content of hippocampus, thalamus and brain stem in goats decreased24%,10%and11%respectively. In midazolam group Asp content of hippocampus and thalamus in goats decreased32%and16%respectively; Glu content of each encephalic region in goats decreased14%,6%,5%,7%and20%respectively. In combined anesthetic group, Asp content of hippocampus and thalamus decreased22%and12%, Glu content of each encephalic region in goats decreased5%,9%,10%,11%and13%respectively.Changes of inhibitory amino acids neurotransmitter in different encephalic regions:In xylazine group, Gly content of thalamus, cerebellum and brain stem increased5%,21%and23%respectively; GABA content of each encephalic region in goats increased60%,19%,74%,92%and75%respectively. In midazolam group, Gly content of hippocampus, thalamus, cerebellum and brain stem increased27%,8%,27%and9%; GABA content of cerebrum, thalamus and brain stem increased59%,36%and58%. In combined anesthetic group, Gly content of hippocampus, thalamus and cerebellum increased23%,20%and33%; GABA content of cerebrum, thalamus, cerebellum and brain stem increased23%,21%,38%and40%.So we can conclude as follows:Hippocampus, thalamus and brain stem are active targets of xylazine on excitatory neurotransmitter in goats; hippocampus and thalamus are active targets of of midazolam on excitatory neurotransmitter in goats; hippocampus and thalamus are active targets of combined anesthetic on excitatory neurotransmitter in goat. There is synergistic effect of xylazine and midazolam on hippocampus and thalamus, and antagonistic effect on brain stem.Thalamus, cerebellum and brain stem are active targets of xylazine on inhibitory neurotransmitter in goats; thalamus and brain stem are active targets of midazolam on inhibitory neurotransmitter in goats; thalamus and cerebellum are active targets of combined anesthetic on inhibitory neurotransmitter in goats. There is synergistic effect of xylazine and midazolam on thalamus, and antagonistic effect on brain stem.
