Objective:The experiment aimde to determine the central nervous mechanisms and effect part of xylazine compound anesthetic and its main components, analyse the rationality of xylazine compound anesthetic, and discuss the effect mechanisms of xylazine compound anesthetic and its main components on different encephalic regions of goats.Methods:65goats were divided into4groups randomly, which intramuscular injected by normal saline, xylazine (12.8mg/kg), midazolam (14mg/kg) and combined anesthetic (1.31mL/kg).The content of NE, DA,5-HT and5-HIAA were determined by high performance liquid chromatography.Results:The variation of catecholamine neurotransmitter of goats in each brain regions:we find that xylazine makes the NE content of each brain regions decrease32.27,35.48%,31.71%,13.11%and7.32%respectively, at the same time, makes the DA content of brain, hippocampus and thalamus decrease33.10%,35.03%and27.01%respectively. Midazolam makes the NE content of brain and brainstem decrease37.71%and31.92%, and the DA content of each brain regions decrease44.80%,11.88%,3.35.%,2.82%and35.81%。 Xylazine combined anesthetic makes the NE content of brain, hippocampus and thalamus decrease42.01%,39.01%and36.71%, and makes the DA content of each brain regions decrease36.61%,42.36%,36.04%,13.35%and8.36%respectively。The variation of serotonin neurotransmitter of goats in each brain regions:we find that xylazine makes the5-HT content of brain, hippocampus and thalamus increase25.90%,44.15%and43.68%respectively, at the same time, makes5-HIAA of each brain regions content increase32.27%,44.60%,49.12%,24.99%and19.97%. Midazolam makes the5-HT content of brain and brainstem increase42.55%and46.58%, and makes the5-HIAA content of each brain regions increase37.19%,6.41%,3.09%,10.36%and51.73%respectively。Xylazine combined anesthetic makes5-HT content of brain, hippocampus and thalamus increase44.52%,56.91%and50.91%, and makes the5-HIAA content of each brain regions increase42.71%,38.79%,52.04%,10.73%and7.96%respectively。Conclusion:To the catecholamine neurotransmitter’s potential active site, xylazine is in hippocampus, thalamus and brain; midazolam is in brainstem and brain; and xylazine combined anesthetic is in hippocampus, thalamus and brain. All these indicate that xylazine and midazolam presents synergistic effect in brain, antagonism in brainstem. To the serotonin neurotransmitter’s potential active site, xylazine is in brain, hippocampus and thalamus; midazolam is in brainstem and brain; and xylazine combined anesthetic is in brain, hippocampus, thalamus. All these indicate that xylazine and midazolam presents synergistic effect in brain, antagonism in brainstem.
