Rapeseed meal is a kind of vegetable protein resources, it must be detoxificatedbefore it is added into feed. Furthermore, there are not only a variety of conventionalnutrients in the leaves of Eucommia ulmoides such as amino acid, vitamin, mineralelements, ect., but also many kinds of unique natural bioactive components such asaucubin, chlorogenic acid, ect.. The technologies of detoxification repeseed meal,extraction and antioxidation activity of phytic acid from detoxification wastewater wereinvestigated. In this dissertation, Eucommia ulmoides and detoxification rapeseed mealwere add into chicken feed. According to trial and error method, chicken were feed andtheir quality was detected, and the effect of the first test was very good. The resultscould provide value-added reliable basis for Eucommia ulmoides and rapeseed meal ofagricultural and forest residues. Main content of this dissertation are summarized asfollows:(1) Study on the technology of detoxification of rapeseed meal by mixed solventThe technologies of detoxification of rapeseed meal were confirmed by singlefactors and orthogonal experiment. According to comprehensive balance analyzing, theoptimum parameters to methanoll-sulfuric acid-water system were as following: takingsulphuric acid (4%) and methyl alcohol (80%) and water (16%) as detoxificationsolvent, liquid-solid ratio8:1(mL/g),40min,45℃,once. As to ethanol-sulfuricacid-water system: sulphuric acid (4%) and ethanol(80%) and water (16%) asdetoxification solvent, liquid-solid ratio8:1(mL/g),40min,45℃,once. Under theseconditions, the removing rate of phytic acid, tannin, glucosinolate, isothiocyanate(ITC)and oxazolidine(OZT) were88.52%,82.43%,92.00%,87.91%and81.65%formethanol system, and were89.34%,83.57%,91.60%,88.32%and82.71%for ethanolsystem,respectively. The results showed that the ethanol-sulfuric acid-water system wasthe better detoxification system than the methanol-sulfuric acid-water system.(2) Preparation of chicken feed with Eucommia ulmoides and detoxificationrapeseed meal as additiveAccording to nutritional needs of chicken and nutrient content of various materials,Eucommia ulmoides and detoxification rapeseed meal of chicken feed was prepared bytrial and error method. The calcium, phosphorus, protein needed during the growth anddevelopment of animals were found. The biological components such as flavonoids,chlorogenic acid and aucubin were found in this feed. Trace element-Se was also foundin the feed, demonstrated it is a kind of rich in Se feed.(3) The effects of chicken feed with Eucommia ulmoedes and detoxificationrapeseed meal on meat performance of chickenThe prepared chicken feed was used to feed chicken and the chicken quality such as slaughter performance, biochemical parameters, conventional indicators andfunctional components were detected. The results show that the feed with Eucommiaulmoides and detoxification rapeseed meal could decrease the food coefficient,abdominal adipose rate, serum cholesterol(CHO), serum triglycerides(TG) and lowdensity lipoprotein(LDL), increase the content of protein and ash. At the same timereduce moisture and fat content of muscle.(4) Study on the extraction and antioxidant activities of phytic acid from detoxifiedwastewater of rapeseed mealPhytic acid extracted from detoxified wastewater of rapeseed meal was inspectedby alkaline precipitation method and its antioxidant activity was assessed in two sides.On the one hand, the acid value and peroxide value of grease on lardy were investigated,on the other hand, the antioxidant capabilities of different free radical of phytic wasinvestigated by chemiluminescence and UV spectrophotometry. The results show thatthe best condition to extract phytic acid was adjust pH8by NaOH. Under this condition,the yield of phytic reached92.83%and the purity was45.96%. Phytic acid had a certaininhibited on rancidity and peroxide of grease. It had the good ability to scavenge everykind of free radical at low density. The IC80of phytic acid extraction on scavenginghydroxyl(·OH), superoxide anion(O2-·), DPPH free radical(DPPH·) and hydroxyl(·OH)were4.20μg/mL,1.99μg/mL,8.99μg/mL and302μg/mL, respectively. So theantioxidant activity of phytic acid extraction was in sequence of O2-·,·OH, H2O2andDPPH·. It was better than Vc except DPPH·. Therefore, the foundation has beendeveloped for natural antioxidant of phytic acid extraction.
