The present most of M. rosenbergiis cultured almost40years in China wereparents`offspring from Japan.This paper regarded Shanghaipopulationafter15yearsbreeding as basic group.Through measuring and processing of basic group dates,finallybody length,the first abdominal segments width and height and carapace width werechoose as measure morphometric attributes.6specialization initial populations were setup by systematic breeding.Each basic group inter-breed, every generation conductselection.44designed primer pairs were screened by useing genomic DNA of3differentpopulations in our courtry, finally the influence factors of getting the number ofpolymorphism of primers were explored.(1) The determine of construction method of M. rosenbergiis specialized strainSpecialized strain is the basis of synthetic strains. Specialized strain which havestable inheritance of the dominant trait is the key of cultivating synthetic strains. Thecultivation of animal husbandry`specialized strain was adopted for. Macrobrachiumrosenbergii through the beginning of the cultivating specialized strain process.It willprovide help at the later stage of the cultivation of M. rosenbergii`specialized strain.Italso provide a reference for the research of other aquatic animals`special line.(2) M. rosenbergii morphometric attributes on body weight in impact analysis391five months old M. rosenbergiis were randomly selected to measure27morphometric attributes,containing body weight,body length,total length,carapacelength, carapace width,carapace,tail ength,number of upper frontal eminencespine,number of lower frontal eminence spine,the length,width and height of eachabdominal segments from the first to the sixth.Simple correlation coefficients andpartial correlation coefficients were calculated. The independent variables which thepartial regression coefficients don`t reach significant level(P<0.05)were eliminated bythe multiple regression analysis of other morphometric attributes as independentvariable and the body weight as dependent variable,and the multiple regression equation of body length,total length,carapace length,carapace height, the first segmentwidth,the fisrt segment height,the second segment width and the sixth segment width tobody weight were established.The principal component regression equation and ridgeregression equation were given by the principal component regression analysis andridge regression analysis on8morphometric attributes retained.The path coefficientsand determinant coefficients of the morphometric attributes on the weight werecalculated.From the result:the simple correlation coefficients of all morphometricattributes to body weight all reach extremely significant level(P<0.01)except number ofupper frontal eminence spine and number of lower frontal eminence spine,but onlypartial correlation coefficients of body length,carapace height,the first segmentwidth,the fisrt segment height and the sixth segment width to body weight achievesignificant level(P<0.05) or extremely significant level(P<0.01).The order of the pathcoefficients value is in line with the determinant coefficients,maximum values are0.387and0.150respectively.A measure guideline about M. rosenbergiis`selecting andbreeding was given.(3) A preliminary study on re-developing of M. rosenbergii microsatellite markers andinfluence factors592M. rosenbergii nucleotides were retrieved in the National Center forBiotechnology Information database,16.39%of the total number of nucleotides containSSRs(simple sequence repeats). Results from the statistics,the dinnucleotide repeat andtrinucleotide repeat are the dominant types,respectively accounting for14.02%and6.08%of nucleotides.29kinds of repeat motifs were discovered from all G-SSRs(Genomic-SSRs).44designed primer pairs were screened by useing genomic DNA ofShanghai population,and32primer pairs showed the distinct amplification,accountingfor72.72%of total primers.Polymorphism detection were commited Jiangsu,Guangxiand Shanghai populations by26pairs of designed primers and37pairs of polymorphicSSR markers from foreign literature,respectively getting9and7pairs polymorphismprimers.The experimental results demonstrate that different populations, sequences ofscreening and primer design directly influence to get the number of polymorphism ofprimers.(4) The structuring of M. rosenbergiis of fast growing specialized strainOn the base of measuring basic group,6specialization initial populations were setup by systematic breeding.The individuals were mating,hatching and raising in eachgroup.When M. rosenbergiis growing,60~90individual were sampled in each groupand measured body length, body weight, carapace height,the first segment width, the firsrt segment height.Processing morphometric attributes`averages and variationcoefficient.
