Red Steppe Cattle is a new national cattle breed cultivated fromMongolian Cattle. It has a lot of excellent traits and is suitable to theclimate of the northeast areas of china. Genetic variation should be takeninto account to direct genetic conservation programs. Microsatellite DNAmarker provides an important role to assess the genetic variation that isnecessary for the Red Steppe Cattle breeding. This research studied the microsatellite polymorphism by Single PrimerAmplification Reaction to the Red Steppe Cattle and its hybrid populationscontaining 1/2,1/4 and 1/8 Limousin blood, and its correlation to dressingcharacters in different hybrid population.The results showed that 8 Single Primer had very good polymorphismin microsatellite amplification. Different primers had different numbers ofbands (5-15) with different sizes mostly between 250bp-2000bp. Similarityfactor analysis was carried out with Microsoft Excel software and thesimilarity factor was between 0-0.5 which suggested that all the 8 singleprime showed very good polymorphism in different hybrid population.With linearity regression analysis and Box-and-Whisker Diagrams, thedifferences of slaughter traits were tested and distribution situation ofdressing traits were analyzed in Red Steppe Cattle and different hybridpopulations. Slaughter traits in 1/4 and 1/2 hybrid populations held obviousadvantages compared with Red Steppe Cattle. Not only in the regressionanalysis but also in Box-and-Whisker Diagrams, the slaughter traits in 1/4and 1/2 hybrid populations had significant differences with pure Red SteppeCattle. The goal of linearity regression analysis and the Box-and-WhiskerDiagrams chart examination is to examine the improvement effects of meattraits, the results showed that breed improvement has already obtained thegood effect during long-term breeding.The relationship between slaughter traits and 8 single primers in RedSteppe Cattle and its hybrid cattle was analyzed with simple correlationanalysis and union canonical correlation analysis. The results of correlationanalysis in different hybrid populations could be used to molecular breedingin future.Anonical correlation analysis has a lot of merits compared to otheranalysis methods. The analysis results showed that primer CAG possessednegative correlation with the body weight, dressing rate, net meat weight,net meat ratio, bone ratio, and body meat production;primer T3B hadnegative correlation with weight daily increasing;primer GTG had positivecorrelation with net meat weight, net meat ratio;primer GACA possessedpositive correlation with fat coverage ratio;primer CGA had positivecorrelation with bone weight and bone ratio.Through the systematic analysis, similarity factors between bands ofmicrosatellite single primers in different hybrid population were compared.Genetic differences were discovered in individuals of Red Steppe Cattleand its hybrid cattle;Differences of dressing traits in Red Steppe Cattle andits hybrid cattle were also analyzed with the significance test andBox-and-Whisker Diagrams, and improvement effects of meat traits wereexamined. The correlation was discovered between mirosatellite singleprimers and slaughter traits. All these results would lay the foundation formolecular breeding.
