Lily is one of the most important ornamental plants in the world. Some great progresses were achieved on cross breeding and germplasm innovation in China, but its molecular marker assisted breeding was behind other staple crops. In this study, the wild endemic triploid species of Lilium lancifolium Thunb. and tetraploid Asiatic hybrids cultivar Brunello as parents were used for distant hybridization to obtain offspring population and to create new germplasm with adaptability, resistance and higher ornamental value. Through the observation of some morphological traits in hybrid progeny, the heterosis and genetic effects of F1 generation of flower and bulbils traits was estimated. Based on the bulbils trait and combined with the BSA method, the AFLP reaction system was optimized and the primers were screened. The main results were as follows:There were some differences in F1 population, which the traits were measured. The variation coefficient changes between 5.39% to 31.06%.The variation of plant height was the most prominent, and 83.78% hybrids was less than their parents and appeared a large ratio of low Pro in plant height. In terms of stem diameter,76.09% of the F1 generation groups were between the parents. The hybrids results showed that only leaf width was close to 1:1:1 in low value parent, mid and high value parent, the other traits, more than 47.83%, were all in the middle of its parents; In terms of floral traits, inner perianth length, inner perianth width, outer perianth length, outer perianth width, flower diameter and number of flowers mid-parent heterosis rate were 4.31%,2.07%,1.67%,-1.40%,15.13% and -43.55%, respectively. Variation coefficient was 5.39%-23.97%. In the aspect of bulbils trait, each group bulbils mean variations of convergence, bulbils number from 1-5 were distributed and extreme changes in the range of 0-17 in hybrid. In F1,22 groups appeared super high parent phenomenon, and nearly 1/4 of the offspring (23.91%,92). No bulbils groups was 11, accounted for 11.96%. Only 5.43% of hybrids were consistent with the female Lilium lancifolium in the number of bulbils, and remaining 68.7% of F1, bulbils number were between their parents.It was appears widely separate in the characters of floral and bulbils, both inner perianth length and flower diameter were more than 1/4 individuals showed transgressive phenomenon, and the ratio of low parent was 0 in the traits of flower diameter and inner perianth width. In addition to outer perianth width and the number of flowers were not shown in heterosis index, the rest were more than 1, indicated that the flower traits of hybrids had heterosis but no heterosis formation. According to the statistical characters of bulbils, mid-value parent was positive and the ratio of heterosis reached more than 30%, compared with the check, F1 generation showed positive in mid parent heterosis, With significant genetic effect and form a certain heterosis in the genetic traits of bulbils.From the changes of kurtosis and skewness, all traits except number of flowers, the absolute value of skewness and kurtosis were less than 3, and showed the continuity of normal distribution, indicated that in addition to the number of flowers all of them were according with a typical quantitative character controlled by multi-genes. The number of flowers kurtosis was 4.190, skewness was 5.762, were more than 3. The kurtosis was excessive, the overall performance was unimodal asymmetric distribution, which was almost in normal distribution deflecting to right. The number of flower characters were quantitative in nature, and controlled by some multi-genes.Genomic DNA was extracted by CTAB method, with a slightly modified. The extracted DNA was showed neat, clear and obscure strip in electrophoresis. Combined with BSA method, based on bulbils trait, analysis of the system optimization of AFLP system and preliminary screening AFLP primers. The results showed that the reaction system was the best while DNA concentration was 30 ngμl-1, dNTPs concentration was 2.0 mmol·L-1, Primer concentration was 1.0 μmol·L-1, Mg2+ concentration was 3.2mmol·L-1 and Taq enzyme concentration was 1.5U, respectively. Through AFLP method 92 materials were analyzed by selected 64 primers, only 29 primers were amplified clear and repeating bands, which got 260 polymorphism bands, with average 8.9 bands each primer that shown a rich genetic diversity and good repeatability among the population.
