Scutellaria baicalensis is a common Chinese herbal medicine, and the mainactive ingredient is flavonoids compounds, including baicalin, baicalein,Wogonoside, Wogonin etc. Secondary metabolites is the major active ingredients inmedicinal plants. Its synthesis level is regulated by plant development and kinds ofbiological or abiotic induced factor. There are several protein in the flavonoidsbiosynthetic pathway to control level of flavonoids such as transcription factors andkey enzymes. In this paper, preliminary explorated the relationship among hormone(gibberellin and abscisic acid), Scutellaria MYB transcription factor genes and therelated genes in flavonoid metabolic pathways by bioinformatics and expressionprofiling analysis.In this paper, results are as follows:(1) There are10R2R3-MYB type and7Single-MYB type MYBtranscription factor candidate genes from Scutellaria baicalensis cDNA libraryused bioinformatics methods, including Blast, GO annotation, InterProScananalysis and phylogenetic analysis. And12MYB transcription factor candidategenes have open reading frame integrity.(2) Subcellular localization analysis showed that SbMYB7and SbMYB8are distributed in the nucleus, while SbMYB2were both distributed in the nucleusand organelles.(3) The transcriptional activity analysis showed that SbMYB2, SbMYB7and SbMYB8have transcriptional activation activity.(4) Relatively expression level analysis showed that the SbMYB5,SbMYB7, SbMYB8, SbMYB12and SbMYB13expression level were a significantchange treatment by ABA, indicating that they may be governed by the ABA.SbMYB2, SbMYB5, SbMYB7and SbMYB8expression were significant changedafter GA3treatment, which may respond to GAs signal, to regulate the downstreamgenes. (5) Constructed a plant expression vector and transformed into Arabidopsisthaliana, T1generation seed was gained.
