Two hundred Hisex layer chicken (1day of age) which were artificially reared to31days of age were randomly divided into4groups (the control group.virus gourpl.virus gourp2, virus gourp3)with50birds in each group. The chicken in the virus groups were challenged by artificial nasal drip according to the results in ELD50of embryos (10-5/0.2ml). The inoculation amount of amniotic fluid with virus in each virus group was10-4/0.2ml,10-5/0.2ml and10-6/0.2ml respectively,0.2mL per bird and the control group was treated with0.2mL sterile saline solution per bird. The contents of CRE, UA, UREA. MDA, SOD, T-AOC and XOD in serum were measured in each group of chicken when they were at17,24and31days old. Five birds in each group were killed randomly for the measurement of the contents of MDA, SOD, T-AOC, XOD in mitochondria extracted from parts of kidney tissues. The remaining of kidney tissues were stablized with10%neutral formalin for the histologically observation and immunohistochemistry. The results were as follow:(1) The productivity changes of chicken infected with kidney type IBV:compared with the control group, the weight gain in virus group1was not significant and the weight of virus group2and virus group3decreased significantly at day17in formal test; At day24, the weight gain in virus group1decreased significantly and that of virus group2and3was not significant; At day31, the weight gain in all of virus groups decreased significantly.(2) The ratios changes between kidney weight and body weight of chick infected with kinday type IBV:at day17, compared with the control group, the difference in left renal/weight, right kidney/weight, bilateral renal/weight were not significant in formal test. At day24, compared with the control group, the difference in left renal/weight, right kidney/weight, bilateral renal/weight of virus group1and gourp3were not significant, while the index in virus group2increased significantly; At day31, compared with the control group, the index in virus group1and group2increased significantly and in virus group3the difference in left renal/weight was not significant but right kidney/weight, bilateral renal/weight increased significantly.(3) renal function changes of chicken infected with IBV:compared with the control group, at day17, the differences of each virus group in CRE, UA and uUREA were not significant in formal test. At day24, compared with the control group, the contents in CRE and UA went up significantly while the difference of content in UREA was no significance in virus group1. The content of UA in both virus group2and group3increased significantly and the difference of content in CRE and UREA were no significance; At day31, compared with the control group, the content of CRE, UA grew significantly and that of UREA was not significant in virus group1. In virus group2. the content of CRE grew significantly and these of UA and UREA were no significance. In virus group3. CRE. UA and UREA contents were not significant.(4) Serous antioxidants and free radical metabolism changes of chicken infected with IBV:compared with the control group, the contents of MDA, XOD in all virus groups were not significant but showed increasing tendency. The contents of SOD, T-AOC were not significant and showed decreasing tendency at day17in formal test. At day31. compared with the control group, the content of MDA grew significantly, that of XOD not significant but slowly increased, and these of SOD. T-AOC not significant but slowly decreased in virus group1. In virus group2. the content of MDA. XOD was not significant but showed increasing tendency and the content of SOD, T-AOC was not significant but showed decreasing tendency. In virus group3. the content of MDA increased significantly, that of XOD was not significant but showed increasing tendency and these of SOD, T-AOC were not significant but showed decreasing tendency.(5) mitochondria antioxidants and free radical metabolism changes of chicken infected with IBV:compared with the control group, the contents of mitochondria MDA. T-AOC and XOD in each virus group were not significant but showed growing tendency and that of SOD grew significantly at day17; At day24, compared with the control group, the content of MDA. SOD increased significantly, that of XOD was not significant but had an increasing tendency and that of T-AOC was not significant but showed decreasing tendency in both virus group1and virus group2. While in virus group3, the content of MDA increased significantly, that of SOD decreased significantly, that of T-AOC had no significant difference but decreasing tendency, that of XOD had no significant difference but increasing tendency. At day31, compared with the control group, the content of the four indexes in each group had no significant difference.(6) Under the observation of light microscopy, in the virus group chicken renal tubular in kidney tissue were expanded and loose, and blood can be found in the connective tissue; renal capsule cavity swelled and bleed sometimes and were filled with uratosis. P53protein immunohistochemical reaction showed that, strong positive signals can be detected in renal tubular epithelial cells in the virus groups, while the control group was negative.
