Effects Of Gelatin And Antioxidants On Boar Spermatoza During Cryopreservation

Compared with almost livestocks, boar spermatoza is easy to be attacked by reactive oxygen species (ROS) in the freezing and thawing process, and the aggregations among spermatozoa lead to the low motility. In order to overcome the negative effects on sperm function after thawing, antioxidants or other substance could be added to boar dilution to prevent oxidative stress and the aggregations.In this study, gelatin and antioxidants were added to frozen dilution. The results indicated that adding200U/mL SOD、0.5mg/mL GSH、0.05mg/mL MLT alone to frozen dilution could increase sperm motility, viability, plasma membrane integrity, acrosome integrity and DNA integrity significantly (P<0.05). Furthermore, the effect of adding combination with100U/mL SOD and0.5mg/mL GSH was more significant than adding antioxidants alone on sperm quality and ability of penetrating zona-free hamster egg (P<0.01).By the way, the gelatin was used in this experiment at the first time and the addition of gelatin to frozen dilution could increase sperm quality and its SPA significantly. Adding0.36mg/mL and0.54mg/mL gelatin could increase sperm motility, viability, DNA integrity significantly and SPA (P<0.01), but no difference between them. Furthermore, there was significant difference on plasma membrane integrity between0.36mg/mL and0.54mg/mL gelatin, and0.36mg/mL gelatin was the best on acrosome integrity.Therefore, addition of0.36mg/mL gelatin or combination with100U/mL SOD and0.5mg/mL GSH to dilution could increase frozen-thawed boar sperm quality and rate of fertilization. At the same time, an effective system of boar semen freezing protection was established to promote the application of frozen semen in the field of artificial fertilization.
