"Active immunity enhancing agent", a combination of many kinds of trace elements (iron, copper, zinc, selenium, manganese, cobal, etc) required by the animal’s body, is the only compound microelement injection of intramuscular in the domestic. It has been proved that "Active immunity enhancing agent" can strengthen metabolism, promote livestock and poultry growth, improve animal performance and enhance the body resistance to diseases, etc. This paper, which took beagle puppy as test object, studied the effect on synergy of "Active immunity enhancing agent" and canine distemper, canine parvovirus bivalent vaccine on the immune function, and lay the foundation for the preparation of new drug declare of the injection and provide the basis to enhance the ability of the pet dogs to prevent and control common infectious diseases. The results were as follows:In the experiment,24health Beagles (6-week-old) were randomly divided into4groups(Ⅰ~Ⅳ groups),2males and2females in each group, given intramuscular injection of Intervet bivalent live vaccine for the first immunization and4-way vaccine for the second and third immunization. Group Ⅰ, vaccine control group, intramuscular injected bivalent vaccine and normal saline of0.2mL/kg body weight,1/d, qd for3d.Group Ⅱ(low dose group), group Ⅲ (middle dose group), group IV (high dose group) for the experimental group, intramuscular injected bivalent vaccine and "Active immunity enhancing agent" of0.1mL/kg,0.2mL/kg,0.4mL/kg body weight, respectively,1/d, qd for3d. Blood samples were drawn at the day before injeeting bivalent live vaccine (reeorded as0d) and after7d,14d,28d,45d.Study on immune enhancement of "Active immunity enhancing agent" to Beagl. The T lymphocyte transformation rate, the antibody level of canine distemper virus and canine parvovirus, the rate of erythrocyte complement receptor rosette (E-C3bRR) and immune complex rosette (E-ICR) were detected. The results show:(1) The lymphocyte transformation rate of the experiment groups was higher than the control. Lymphocyte transformation capacity of high-dose group was extremely significant higher during the experiment (P<0.01). The rates of group Ⅲ and Ⅱ were significant or extremely significant lower than group Ⅳ respectively from28days, while there was no marked difference between the two groups.(2) The antibody of CDV and CPV of the experiment groups were generated rapidly and the antibody level was increased after intramuscular injecting "Active immunity enhancing agent". The CPV antibodies in serum of the experiment groups were positive at7th day after the first immunization and the level was extremely significant increased compared with that in the control group (P<0.01). The CDV antibody levels were very significantly higher than the control from28days. The result showed "Active immunity enhancing agent" might strengthen the humoral immune response, the high and middle dose group the best were better (P>0.05).(3) The E-C3bRR percentages of the experiment groups were increased in varying degrees and the rate of E-ICR was reduced. Comparing with the control group, The E-C3bRR percentages of group Ⅳ and Ⅲ were extremely significant higher and the E-ICR percentages were significant or extremely significant lower from14days. The result showed erythrocyte immune function could be improved.Analyze the effect on antioxidant indicator and related cytokines of "Active immunity enhancing agent" to immune Beagles, GSH-PX and SOD activity, MDA, IL-1, IL-2and IL-6levels, ant-hydroxy radical capability in serum were determined. The results show:(1) The activity of serum GSH-PX and SOD, the ant-hydroxy radical capability were significant or extremely significant increased in the middle dosages and high dosages group, while the content of MDA was decreased extremely significant, indicating that a certain dose of "Active immunity enhancing agent" could enhance the antioxidant capacity.(2)"Active immunity enhancing agent" was able to increase the contents of IL-1, IL-2, IL-6in serum. Compared with the control group, the high dose group showed significant or extremely significant difference (P<0.05or P<0.01). It showed the immune enhancement of "Active immunity enhancing agent" to Beagl was related with the secretion of IL-1, IL-2, IL-6. Conclusion:"Active immunity enhancing agent" synergy with canine vaccine can promote the immune performance, improve T lymphocyte transformation rate and the antibody level of CDV, CPV in serum, enhance immune function, improve the antioxidant capacity, promote the secretion of IL-1, IL-2, IL-6in serum, and " Active immunity enhancing agent" used in middle dose (0.2mL/kg body weight) is better and feasible in clinical.
