A long-term survey shows that bariatric surgery can not only control blood sugar in patients with type2diabetes, but also prevent obesity and impaired glucose tolerance progressing to type2diabetes, and reduce the morbidity and mortality of patients with type2diabetes.Many research results show that bariatric surgery can significantly improve the obesity patient’s blood glucose and impaired glucose tolerance in patients with type2diabetes. A considerable number of patients do not need drugs hypoglycemic and maintain normal blood sugar chronically.Bariatric surgery is a good treatment for obesity and type2diabetes,and has extensive clinical application at home and abroad.Surgical research has been carrying out in-depth development, but its therapeutic mechanism and side effects on the body is studied by few people.Bama miniature pig has a great similarity with the human anatomical structure, physiological functions, and intestinal flora.It is considered to be a suitable animal model for assessing human gut microflora diseases.Objective The most mature surgical bariatric surgery Roux-en-Y gastric bypass(GYBP) was token in the study.Through the adult Bama miniature pigs after GYBP,we studied serum markers and changes in intestinal flora and got the actual impact on body for preliminarily assessing the security of gastric bypass surgery,which provided the basis for Bama miniature pig to be the animal models on gut microflora.Method After GYBP,6female mature Bama miniature pigs were fasted drink and eating for three days, during which1500ml glucose and1500ml saline were infused every pig per day.Then water and feed the liquid food (milk,750ml per day).7days later,give the normal diet.Collect blood of surgery and7months later.After4hours at room temperature,centrifuged3000rpm for5minutes.Stored the serum at-20℃Collect feces of normal,1month later,2months later,3months, sealed by anhydrous ethanol, also placed in a-20℃.Finally do determination of the total DNA of the fecal flora, the16SrRNA gene V3region sequences,denaturing gradient gel electrophoresis (DGGE). Then analyze serological markers.Result The richness (s) of normal group, surgery after one month group, surgery after two month group and surgery after three month group were6.44±0.33,5.30±1.50,7.22±0.60and7.47±0.76; the Shannon Index (H’)of them were3.29±0.05,3.12±0.34,3.42±0.11,3.40±0.21respectively; the Similarity Dice between normal group, surgery after one month group and surgery after two month group, surgery after three month group was63%.In the statistical analysis of the serological, there are two significant differences on creatinine and total bilirubin.Conclusion Gastric bypass surgery caused some impact on the gut micro flora of model pigs, but not significant.In addition, the surgery may damage the liver, and reduce glomerular pass rate.
