The Effects Of Different Diets On Cytokine And Hormone Level In Landrace And Bama Miniature Pigs

In order to reveal the immunology basics of different disease resistance between local and foreign pig breeds and the molecular mechanism in growth hormone regulating growth and development, this test,using the5week old Landrace and Bama miniature pigs as objects of study, sampled after feeding in three divided stages in two different nutrient level conditions and slaughtering, then measured serum cytokine and hormone levels of two types of pigs,and then analyzed the levels of cytokine mRNA expression in jejunum, spleen, muscle and liver hormone receptor mRNA expression in muscle and liver. The results are as follows:1、Serum concentration of cytokine and expression level of tissue mRNAIn phase one,the same pig breeds’serum concentration of TNF-a,the local diet group’s is higher than the NRC diet group’s(P<0.05);In phase three,the Bama miniature pig serum concentration of IL-2,the NRC diet group’s is higher than all the other groups’(P<0.05);In all the three phrases,the Bama miniature pig serum concentration of IL-10,the local diet group’s is higher than all the other groups’(.P<0.05);In phrase two,the jejunum IL-4mRNA expression level in same diets,Bama miniature pigs’is significantly higher than Landrace pigs’(P<0.05);In phrase three,the jejunum IL-6、 IL-10(phrase two’s is contrary)、IL-1β、IFN-y(including phrase one and phrase two)、TNF-α and TGF-β1(including phrase two)mRNA expressions are all that Landrace pigs’is higher than Landrace pigs’in the same diets(P<0.05);In phase one,spleen IL-2、IL-6(including phase two and phase three)、IL-10(including phase three but phase two’s is contrary)and TGF-β1mRNA expressions are all that Landrace pigs’is significantly higher than Bama miniature pigs’between the same diet teams(P<0.05).2、Serum hormone level and corresponding tissue receptor mRNA expression levelIn phrase one,between same diet teams,Bama miniature pigs’serum GH level is significantly higher than Landrace pig team’s(P<0.05);in three phrases,serum IGF-1levels are all that Landrace pigs’is significantly higher than Bama miniature pigs’;in phrase three,Bama miniature pigs’ serum SS level is significantly higher than Landrace pigs’(P<0.05);In the same diet teams,The longissimus muscle GHR (phrase one) and IGF-2(including three phrases) mRNA expression levels are all that Landrace pigs’is significantly higher than Bama miniature pigs’(P<0.05);in phrase two,IGF-1R and IGF-2R expression levels are all that Landrace pigs in local diet team’s are significantly below all the other teams’(P<0.05);in phrase two IGFBP-3and IGFBP-5mRNA expression levels are that Bama miniature pigs’is significantly higher than Landrace pigs’(P<0.05);in phrase two and three(phrase one’s is contrary),Bama miniature pigs’TGF-β1mRNA expression level is significantly higher than Landrace pigs’(P<0.05);In phrase one,femoral biceps GHR、IGF-1、IGF-1R、IGF-2R、IGFBP-3、IGFBP-5and TGF-β1mRNA expression levels are that Bama miniature pigs’is significantly higher than Landrace pigs’between the same diet teams(P<0.05);in phrase two,IGF-1R、IGFBP-3and TGF-β1mRNA expression levels are contrary to phrase one’s;in the three phrases,IGF-2mRNA expression levels are all that Landrace pigs’is significantly higher than Bama miniature pigs’(P<0.05);In phrase two,GHR and IGF-1R(the phrase three’s is contrary)mRNA expressions in liver are that Bama miniature pigs’is significantly higher than Landrace pigs’between the same diet teams(P<0.05);phrase one’s IGF-1、phrase two’s and phrase three’s IGFBP-3and phrase three’s TGF-β1mRNA expression levels are all that Landrace pigs’is significantly higher than Bama miniature pigs’(P<0.05).The experimental results showed that different pig feeds’ cytokines serum concentration and the corresponding organization mRNA expression level、serum hormone level and the corresponding organization receptor mRNA expression level are different, and different diets can also affect the results.
