To evaluate the mechanisms and the effects of right dose of cabergoline on the fertility of mice, which are useful for developing the safe and effective rodent sterilant, cabergoline was given by gastrogavage during different periods of mice, and the results were as following:One dose of cabergoline treatment on mature female mice had no effect on the reproductive rate, litter size at birth and the growth of the pups, however, the survival rate of pups was significantly decreased. The death litter number of150μg/kg、300μg/kg、600μg/kg groups were2,7,7respectively; the mortality number of pups were4,10,12respectively; the survival rate of pups were96.85%、92.59%、91.24%; while the pups of controls had no death. This study indicated that one dose of cabergoline on mature female mice could influence the survival of their pups.200μg/kg Cabergoline treatment at1,2, and3days of gestation could effectively control the reproduction of mice.100μg/kg Cabergoline treatment at1,2, and3days of gestation could reduce the reproductive rate, litter size and survival rate of pups, and the reproductive rate, litter size and survival rate of pups were50%,72%,89%in comparison with the controls, but the development of pups in100μg/kg group was significantly greater than the controls.200μg/kg or400μ/kg Cabergoline treatment at1,2, and3days of gestation could completely inhibit the reproduction of mice, and the fertility did not fully restore after the second mating (44%,40%respectively); but the reproduction indexes between200μg/kg and400μg/kg had no significantly difference. These demonstrate that200μg/kg Cabergoline treatment at1,2, and3days of gestation can effectively control the reproduction of mice.Cabergoline treatment on15,16,17days of gestation did not impact the reproductive rate and litter size at birth, while the litter size at weaning and survival rate of pups were remarkable decreased. The mortality numbers of50μg/kg,100μg/kg,200μg/kg,400μg/kg groups were12,14,20,32, the survival rate were91.61%,90.07%,84.62%,76.81%, at the same time, the mortality number of pups and the dose of cabergoline were positively related, moreover, the growth of pups between the cabergoline treatment groups and the control had no significantly difference. These results showed that cabergoline treatment during late pregnancy of mice could effectively decrease the survival rate of pups.Cabergoline treatment on1,2,3days of lactation could significantly reduce the survival rate of pups, and significantly inhibited the development of pups for up to7days. The litter size at weaning and survival rate of pups in50μg/kg,100μg/kg,200μg/kg, and400μg/kg groups were significantly lower than the controls, survival rate of pups were96.15%,95.92%,92.62%,78.81%respectively. The study indicated that cabergoline treatment during early lactation could effectively decrease the survival rate of pups, and transitorily inhibit the development of pups.In conclusion,0.8ml400μg/kg cabergoline was the minimal dose to inhibit the reproduction of mice during pregnancy and lactation periods. The mechanism of this dose of cabergoline to inhibit early pregnancy was to prevent the implantation of fertilized ovum and terminate the pregnancy; the mechanisms to control the late pregnancy and early lactation were to suppress the milk secretion, thus, the pups were partially died due to malnutrition, further, the survival rate of pups was reduced. These results pointed out that moderate dose and continuous throw of cabergoline bait should be adopted, hence, mice pest would ingest bait continuously and reach the best effects; in addition, the infertility control would be better if we offered bait during the breeding season.
