Pyrus pyrifolia ’Hosui’ was used as the material to study in vitro culture of stem segments and shoot-tips and plant regeneration. The optimal medium for in vitro culture of stem segments was LP+ IBA 0.4mg/L +6-BA 0.5mg/L +VB5 1.0mg/L +VH 0.1mg/L +VB2 0.5mg/L+VC 1.0mg/L. The optimal subculture medium was LP+ IBA 0.4mg/L +6-BA 0.5mg/L VB5 1.0mg/L +VH 0.1mg/L +VB2 0.5mg/L. On this basis, the shoot-tips in vitro culture and rooting culture were mainly experimented. The main results were as follows:1. The optimal IBA concentration for shoot-tips in vitro culture of Pyrus pyrifolia ’Hosui’ was between 0.5mg/L and 1.0mg/L.2. The optimal basic medium for rooting culture in vitro of Pyrus pyrifolia ’Hosui’ was l/4MS.The rate of it is 68.75%.3. Based on 1/4MS as basic medium, the optimal IBA concentration for rooting culture in vitro of Pyrus pyrifolia ’Hosui’ was 0.3 mg/L. The rate of it is 75.0%.4. Based on 1/4MS as basic medium, the optimal PG concentration for rooting culture in vitro of Pyrus pyrifolia ’Hosui’ was 80.0 mg/L. The rate of it is 87.5%.5. Dark culture was essential to rooting culture in vitro of Pyrus pyrifolia ’Hosui’. The optimal days of dark culture was 7 to 9 days.6. Before the plantlets were transplanted, the roots were washed clearly and dipped in nutritive medium for 7 days, and then transplanted to the matrix with vermiculite and fertilized soil (1: 1)...
