Fowl coccidiosis is one of the most common parasitic protozoa diseases in poultry, which is a most serious hazard of fowl parasitosis. The disease incidence rate ranged from 30% to 100%, while the mortality rate maybe appeared from 20% to 80%. The growth rate of those chickens suffering this disease is significantly slow. Fowl coccidiosis is a kind peroral infection disease and the oocyst is the infection sources, killing the oocyst may prevent this disease completely. However, oocyst has the power of resistance to the common used disinfector and the surrounding environment. According to the own traits of the coccidian oocyte, we developed the oo-stat, which is aim directly at the coccidian oocyte.Via selected the designed different oo-stat formula, this project studied oo-stat killed the oocysts (both spored and unspored) from in vivo and in vitro aspect, and the effection from the outer environment on it. Finally, we carried on the clinical trial of oo-stat in real cultural environment, the concrete content are as follows:1 Study on optimized trial of oo-stat formulaThis experiment was conducted to obtain the best oo-stat formula this inhibited and killed coccidian oocyst. We could get different concentrations 1:20,1:30,1:40,1:50,1:60, 1:80,1:100,1:150,1:200,1:300 and 1:400 from three different formulas of oo-stat, and observed their spored-ratios after E.tenella unspored-oocyst were interacted by different concentrations 6h,12h and 24h. The results showed that spored-ratios were significantly lower than negative control group (P<0.05) except the concentrations of 1:300 and 1:400 interacted 6h and 1:400 interacted 12h of oo-stat formulaⅠand formulaⅡ. E.tenella unspored-oocyst were treated by concentrations of 1:20,1:30,1:40,1:50,1:60 and 1:80 of oo-stat formulaⅠandⅡafter 6h,12h and 24h, the spored-ratios were lower than 5%; and if E.tenella unspored-oocyst were treated by oo-stat formulaⅢin the concentrations of 1:20, 1:30,1:40,1:50,1:60 and 1:80 after 12h and 24h, the spored-ratios were also lower than 5%, but it were treated 6h, we found spored-ratios were quite higher. These results indicated that oo-stat formulaⅡhas excellent capability to inhibit and kill coccidian oocyst. 2 Effect of centigrade on the oo-stat to inhibit and kill fowl coccidian unspored-oocystThis experiment was conducted to study the effect of centigrade on the oo-stat to inhibit and kill fowl coccidial unspored-oocyst,7% ammonia water was used as the control group and oo-stat of 1:50,1:100,1:150 were employed as the solution groups, while the normal saline were adopted as the positive control group. These solutions interacted with E.tenella unspored-oocysts for 6 h,12 h,24 h in vitro,2.5% K2Gr2O7 solution was used to cultivate the coccidian oocysts.Finally, the spored-ratios were obtained. The results showed that the spored-ratios at 4℃were significantly higher than relevant treat groups dealed with 20℃under the oo-stat of 1:50,1:100,1:150 (P<0.01). These results indicated that the oo-stat has a better effect on inhibit the spore of the coccidian oocyst at the twenty centigrade.3 Study on oo-stat inhibit and kill spored-oocyst of fowl coccidiaThis experiment was conducted to observe the effect of oo-stat on inhibit and kill fowl coccidial spored-oocyst.7% ammonia water was used as the control group, and the oo-stat solution of 1:50,1:100 and 1:150 dealed with spored-oocyst for 6h,12h, and 24h, respectively. Then ratio was 5×104 treated coccidian oocysts to one fowl to inoculate, and set the positive control group of inoculating untreated oocysts and the negative control group of uninoculating coccidian oocysts. After observing the clinical behaviours and lesions of each inoculated fowl,the anti-coccidia index was calculated. The results showed that the anti-coccidia effection of the oo-stat of 1:50 at various time treat group were highest performence; the oo-stat groups of 1:100 treated 12h,24h were medium; the oo-stat groups of 1:150 displayed the lowest, even ineffective. These results indicated that the oo-stat solution of 1:50 demonstrated strong force at kill coccidian spored-oocyst.4 Effect of organics on the oo-stat to inhibit and kill fowl coccidian oocystThis experiment was conducted to study the effect of organics on oo-stat in fowl coccidian oocyst. Through using the oo-stat solution of 1:50 under 20℃,coccidian oocyst and the mixture of oocyst and feces (spored-oocyst and unspored-oocyst) were treated respectively, then observing information of the growth and lesions of the fowls of inoculated the spored-oocyst and by checking the spored-ratios of the oocyst to measure the effect of organics on oo-stat. The results showed that the spored-ratios of the oocyst were higher when the feces existed and the effect of oo-stat was attenuated, as the high adding of the fowl feces. When the feces existed,oo-stat almost had no effect on the coccidian spored-oocyst, and the mortality of the inoculated fowls is higher than control group. These results indicated that organics has certain impact to the oo-stat on the inhibiting and killing fowl coccidian oocyst.5 Imitation field experiment of oo-stat inhibit and kill fowl coccidian oocystThis experiment was conducted to study the effect of the oo-stat on inhibit and kill fowl coccidian oocyst under imitation field trial. The floor of henhouse that sprayed with the mixture of different species fowl coccidian spored-oocyst was conspergered with the oo-stat solution of 1:50 with the doors and windows closed. The fowls were put in the henhouse by groups after dealed with 24 h. The positive control groups and negative control groups were set, and repeated the same treatment twice. The results showed that the oo-stat has an excellent effect compared with the positive control groups, the average weight gain of experiment groups were extreme significance higher (P<0.01),and relative weight gain rate were higher. The feed conversion rates of experiment groups were higher than positive control groups. These results indicated that the oo-stat has an excellent capability on inhibit and kill fowl coccidian oocyst in imitation field trial.6 The field trial of oo-stat preventing fowl coccidiosisThis trial to gain the proper concentration of the clinical used oo-stat. Those hen houses which suffered great coccidiosis were selected to do trial and were disinfected with oo-stat of 1:25,1:50 and 1:100, respectively, then closed the doors and windows for 24h. No using of oo-stat was as control groups and repeated the same treatment twice. The results showed that the feed conversion ratios of the oo-stat treatment groups were lower than positive control groups, and EEF of the oo-stat groups were higher than control groups. The mortality of the different concentration were lower than the control group; the treatment of 1:25 and 1:50 were tested a few oocysts at padding, only. The lesions of the treatment of the different concentration were lower than the mean value and control group is higher than it. These results indicated that oo-stat of 1:50 has the better killing effect.
